Abstract: Ziel dieser Arbeit war die Entwicklung von Matrix-Retardgranulaten mit Hilfe des Walzenkompaktors, Mini-Paktors® der Firma Gerteis, sowie die Untersuchung des Einflusses von Rezepturveränderungen und Variationen der Kompaktierparameter auf die Freisetzung. Ein Vorteil der eingesetzten nichtquellenden Hilfsstoffe ist die Möglichkeit zur Weiter-verarbeitung zur Multiple-Unit-Form. Zudem bieten die so hergestellten Matrix-Retardgranulate mit einem Durchmesser unter 2 mm den Vorteil, dass sie bei geschlossenem Sphinkter den Pylorus passieren können, so dass die Verweilzeit im Magen erheblich verkürzt wird. Eingesetzt wurden die Matrixbildner Ethylcellulose 7cP und 100cP, Compritol® 888 ATO und Precirol® ATO 5 und als Modellarzneistoff Theopyhllin-Monohydrat. Auswirkungen durch Veränderung der spezifischen Kompaktierkraft, der Granuliersiebmaschenweite und der Rezeptur der hergestellten Granulate, besonders des Arzneistoffgehalts, auf die Freisetzungen wurden beobachtet. Dabei kann man verallgemeinern, dass eine höhere Kompaktierkraft, ein gröberes Granuliersieb und ein geringerer Arzneistoffanteil die Freisetzungsgeschwindigkeit des Arzneistoffes aus dem Granulat verringert. Die Granulate mit Ethylcellulose 100cP wiesen keine retardierenden Freisetzungseigenschaften des Wirkstoffes auf. Weiter konnte gezeigt werden, dass durch Zugabe von weiteren Hilfsstoffen, insbesondere Kollidon® SR, zum Matrixbildner Precirol die Freisetzungsprofile stark modifiziert werden können. Das lässt eine größere Variation der Wirkstofffreisetzung zu.
The aim of this study was the develop of non-swellable matrix sustained release granules by roll compaction, Mini-Pactor® of Gerteis, and the investigation the variation of ingredients and the roll-compaction parameters of the dissolution. An advantage of the non-swellable excipients is the manufacture to a multiple-unit-from. Matrix sustained release granules under a diameter of 2 mm pass the stomach with the close Sphincter, to reduce the dwell time. The used matrix former are ethyl cellulose 7cP and 100 cP, Compritol® 888 ATO and Precirol® ATO 5 and as model drug theophylline monohydrate. Observed the dissolution by variation of specific compaction force, the granule sieve-size and the ingredients, the amount of drug content, of the produce granules. Generally with a higher specific compaction force, a coarse granule sieve-size and a low amount of drug, reduce the dissolution speed. Ethyl cellulose 100cP are not suitable to made a granules with sustained release dissolution characteristic. It showed that the addition of other excipients, especially Kollidon® SR, to the matrix-former Precirol® ATO 5 can intense mode the dissolution profile of the produce granules. That can be used for a greater range of drug dissolution.
