Abstract: Seit Jahrzenten gehören Actinomyceten und vor allem das Genus Streptomyces zu den wichtigsten Produzenten von biologisch aktiven Naturstoffen. Mittels einer bioaktivitätsgeleiteten Herangehensweise sowie HPLC-UV und MS basierten Methoden, wurde ein neues Hydrazonderivat der Aminosäure Leucin aus dem Ethylacetatextrakt des Stammes Streptomyces fradiae Tü 27 isoliert. Neben einer geringen Zytotoxizität des Hydrazons, waren wir in der Lage eine erhöhte Zytotoxizität gegen HeLa-Zellen zu beobachten, wenn das Hydrazonderivat mit den zytotoxischen Aminoglycosidantibiotika Hygromycin B und Geneticin kombiniert wurde. Die Bestimmung des „Fractional inhibitory concentration index“ ergab eine additive Wirkung für die Kombination mit Hygromycin B. Zudem wurden bakterielle Endophyten aus marokkanischen Makroalgen isoliert. Die Anwendung der „One strain-many compounds“ Methode sowie MS und MS/MS basierten Analysen ergab die Produktion der sogenannten Globomycine für alle isolierten Endophyten.
Actinomycetes and especially the genus Streptomyces have been an important source for biologically active specialized metabolites for decades. Using a biological activity guided workflow as well as HPLC-UV and MS based analyses, a novel hydrazone derivative of the amino acid leucine was isolated from the ethyl acetate medium extract of the Streptomyces fradiae strain Tü 27. Beside a low cytotoxic activity of the compound itself, an increase in cytotoxicity against HeLa cells was observed when the compound was combined with the cytotoxic aminoglycoside antibiotics hygromycin B, and geneticin. The calculation of the “Fractional inhibitory concentration index” revealed an additive effect for the combination with hygromycin B. Furthermore, bacterial endophytes have been isolated from different macroalgae of Moroccan origin. Using the “One strain-many compounds” approach as well as MS and MS/MS based analyses, we proved the production of different globomycins for these endophytes.
