Abstract: Die Arabidopsis thaliana Gene At1g73010 und At1g17710 sind Phosphatmangel-induzierte Gene und kodieren für Phosphatasen der HAD-Superfamilie. Während das Gen At1g73010 für eine Pyrophosphat-spezifische Phosphatase kodiert, repräsentiert das andere Gen die erste Phosphoethanolamin (PEA) und Phosphocholin (PCho)-Phosphatase in Pflanzen, benannt als AtPECP1. Die aktuelle Studie zeigt das das Mg2+-abhängige Enzym hoch spezifisch für beide Substrate ist und ein enzymatischkatalysiertes Optimum im sauren bis neutralen Bereich besitzt. Der apparente Km-Wert für PCho beträgt 0.44 mM und ist niedriger als der Km-Wert für PEA mit 1.38 mM, jedoch ist die katalytische Effizienz für PEA deutlich charakteristischer. Gelfiltrationsstudien zeigten, dass die zytoplasmatische PECP1 als Monomer und Dimer existiert und in einer schnellen Gleichgewichtsreaktion zwischen beiden Formen enzymatisch aktiv. Die Arabidopsis-Proteine sind homolog zu einer Anzahl von HADGenen anderer Pflanzenspezies. Alle Phosphatasen kodieren für Phosphoethanolamin/-Phosphocholin-Phosphatasen und bilden die neue PECP-Enzymfamilie in Pflanzen.
The Arabidopsis thaliana genes At1g73010 and At1g17710 are phosphate-starvation introduced genes and encoded for phosphatases of the HAD superfamily. While the At1g73010 gene determined for a pyrophosphate specific phosphatase the other gene preset the first phosphoethanolamine (PEA) and phosphocholine (PCho) phosphatase in plant, named AtPECP1. This study show that the Mg2+-dependent enzyme exhibits a high specific activity for both substrate and an optimal enzymatic catalysis at acid to neutral range. The apparent Km value for PCho is with 0.44 mM lower as the Km value for PEA with 1.38 mM but the catalytic efficient for the substrate PEA is distinct promoted. Gelfiltration revealed the monomer and the dimer structure of the soluble cytoplasmic protein, which active catalyze the cleavage of the substrates independent of each other. Arabidopsis proteins are homologous to a set of genes from other plants species that were characterized so far with respect to Pi-starvation. Surprisingly, the homologous phosphatases are homologue enzymes to the first Arabidopsis Phosphoethanolamine/Phosphocholine-Phosphatase1 (AtPECP1) in plants.
