Abstract: Streptomyces coelicolor A3(2) ist ein gram-positives, obligat aerob wachsendes Bodenbakterium mit den charakteristischen Merkmalen einer differenzierten Entwicklung (vegetatives Substratmyzel, hydrophobes Luftmyzel, Sporen) und einer ausgeprägten extrazellulären Biologie, die die Produktion vielfältiger Sekundärmetabolite (u.a. Antibiotika) einschließt. Trotz seiner ausschließlich als obligat aerob charakterisierter saprophytischen Lebensweise kann S. coelicolor A3(2) plötzlich auftretende anoxische Bedingungen sowohl als Spore als auch Myzel wochenlang überdauern. Einen möglicherweise wichtigen Anteil für diese Fähigkeit bieten drei im Genom annotierte putative respiratorische Nitratreduktasen (Nar) und damit eine möglicherweise anaerobe Atmung mit Nitrat. Die Dissertation zeigt unter Verwendung neu entwickelter Methoden, in welchen Entwicklungsphasen und unter welchen Bedingungen die unterschiedlichen Nar-Enzyme von S. coelicolor A3(2) synthetisiert werden und eine Aktivität zeigen.
Streptomyces coelicolor A3(2) is a soil-dwelling, obligate aerobic high GC Gram-positive bacterium with a complex life cycle including three different stages (substrate mycelium, aerial mycelium and spores). Moreover it is known for a well-marked extracellular biology – especially its secondary metabolite production (e.g. antibiotics). Remarkably for an obligate aerobe, S. coelicolor A3(2) survive a couple of weeks under anoxic conditions as spore as well as mycelium. Due to three putative respiratory nitrate reductases (Nar), which were encoded in the genome, it seemed to be possible that an anaerobic respiration with nitrate contributes the anaerobic survival. In this dissertation it is shown for all three Nar-enzymes in which developmental stages and under which conditions they are synthesized and active. Therefore new developmented methods were used.
