Abstract: Diese Arbeit behandelt die Charakterisierung der nicht-kodierenden Y RNAs. Es wurden RNA pulldowns durchgeführt, um Y RNA-assoziierte Proteine zu identifizieren (z.B. das IGF2 mRNA-bindingprotein 1). Des Weiteren wurde gezeigt, dass mRNA-Prozessierungsfaktoren mit Y RNAs interagieren. Die Depletion von Y RNAs mittels antisense oligonucleotides (ASOs) verursachte Histone-spezifische Prozessierungsdefekte. Y3’s Rolle in der 3’-Endprozessierung ist nicht konserviert in der Maus, welches im Fehlen der kleineren Y3-Form Y3** begründet liegt. ASO- vs. siRNA-vermittelte Y3**-Depletion bestätigte die Rolle dieser RNA in der Histon-Prozessierung. Während Y3 und Y3** mit Prozessierungsfaktoren interagieren, kann nur Y3** mit Histon-mRNAs assoziieren. Außerdem beeinträchtigte die Depletion von Y3** die Morphologie/Dynamik von histonelocusbodies (HLBs). Zusammenfassend konnte gezeigt werden, dass Y3** Histone-mRNAProzessierung fördert, indem sie Prozessierungsfaktoren zu HLBs rekrutiert.
This thesis focused on the characterization of non-coding RNAs called Y RNAs.To identify Y RNA-associated proteins we performed RNA pulldowns. This identified Y RNA-binding proteinsincluding the IGF2 mRNA-binding protein 1. Furthermore we identified the association of mRNA processing factors with Y RNAs. Y RNA depletion by antisense oligonucleotides (ASOs) caused processing defectsspecific for histone mRNAs. Y3’s role in the 3’-end processing of histone mRNAs is not conserved in mouse, which is caused by the lack of a smaller Y3-variant, termed Y3**.ASO- vs. siRNA-directed depletion of Y3** revealed Y3** to be involved in histone mRNA processing. Furthermore,while Y3 and Y3**associate with processing factors, just Y3**associates with histone mRNAs. The depletion of Y3** impaired the morphology/dynamics of histone locus bodies (HLBs). In conclusion, these findings indicate that Y3** promotes processing of histone mRNAs acingas scaffold to recruit processing factors to HLBs.
