Abstract: Die RhoGTPasen Cdc42 und Rac1 spielen eine wichtige Rolle in der Knochenentwicklung. Mit Hilfe eines Mausmodells wurden Cdc42 und/oder Rac1 in Präosteoblasten ausgeschaltet. Die konditionellen Knockout (cKO)-Mäuse sind kleiner als die CT (Kontroll)-Tiere. Die Knochendichten waren in allen cKO-Gruppen vermindert. Histomorphometrische Analysen zeigten, dass die Ausbildung der sekundären Spongiosa beeinträchtigt ist. Dies war in allen cKO-Tieren auf eine reduzierte Mineralisierungsfähigkeit zurückzuführen. Versuche mit isolierten Calvaria-Präosteoblasten bestätigten diese Ergebnisse. Zudem war die Adipozytenanzahl im Knochenmark der cKO-Mäuse erhöht, verglichen mit der der CT-Tiere. Adipozyten leiten sich wie Osteoblasten von mesenchymalen Stammzellen ab. Unsere Ergebnisse deuten auf eine Beteiligung der Rho GTPasen an Signalwegen, die die osteogene Entwicklung fördern und die Differenzierung zu Adipozyten unterbinden, hin. Durch den Verlust eines der Faktoren wird diese Kaskade gestört.
The RhoGTPases Cdc42 and Rac1 are involved in bone development. With the help of the Osterix-promotor Cdc42 and/or Rac1 were deleted in preosteoblasts. The conditional knockout (cKO) mice were much smaller than their littermates. Also the bone density was decreased in all cKO-samples. Histomorphometric analysis showed that the development of the spongiosa was affected by the loss of Cdc42 and/or Rac1. All cKO-mice showed a reduced ability of mineralization. Furthermore we found a high number of adipocytes in the bone marrow of cKO-mice. Adipocytes and osteoblasts evolve from the same mesenchymal stem cells. Our results let us postulate a transdifferentiation of preosteoblasts into adipocytes. Cdc42 and Rac1 seem to play a key role in bone development and differentiation of mesenchymal stem cells. The loss of one of the investigated Rho GTPases disturbs the formation and function of osteoblasts.
