Abstract: Sowohl Fett- als auch Skelettmuskelgewebe werden als wichtige sekretorische Organe angesehen. Die von ihren Zellen gebildeten und freigesetzten Proteine, sogenannte Adipo- bzw. Myokine, können auto-, para- und endokrine Wirkungen entfalten und darüber kommunizieren. Bezüglich ihrer physiologischen Funktionen bei Rindern sind bisher kaum Informationen vorhanden. Die vorliegende Arbeit verfolgte daher das Ziel, potentielle Moleküle zu finden, die den Muskel- und Fettstoffwechsel von Mensch, Modelltieren und Rindern beeinflussen. Über eine umfangreiche speziesübergreifende Datenanalyse konnten putative Kandidatengene identifiziert werden. Ausgewählte Faktoren, die für die Produktqualität und Tiergesundheit beim Rind von Bedeutung sein könnten, wurden initial charakterisiert. Durch die Etablierung von Zellkulturmodellen wurden methodische Voraussetzungen geschaffen, um den Einfluss bestimmter Zytokine auf das Wachstums- und Differenzierungsverhalten von bovinen Primärzellen zu untersuchen.
Adipose tissue and skeletal muscle are recognized to be important secretory organs. Both tissues produce and release proteins, so-called adipokines and myokines, enabling them to exhibit auto-, para- and endocrine activity and communicate with their local environment. In cattle, the information existing about their physiological function is rather limited. The present work aims at the identification of potential molecules influencing the cross-talk between adipose tissue and skeletal muscle in humans, laboratory rodents and cattle. In a wide comparative cross-species analysis of differently regulated transcripts putative candidate genes could be identified. Selected factors, which might be relevant for product quality and animal welfare of cattle, were initially characterized. With the establishment of cell culture models, additional methodological preconditions were created to investigate the effect of special cytokines on the proliferation and differentiation of bovine primary cells.
