Abstract: Durch Ableitung von Struktur-Wirkungs-Beziehungen bekannter MDR-Modulatoren wurden neue, potentielle Inhibitoren synthetisiert. Diese wurden durch biochemische in vitro-Untersuchungen an verschiedenen Krebszelllinien bezüglich ihres inhibitorischen Potentials an P-Glykoprotein, eine der bedeutendsten Effluxpumpen im Zusammenhang mit Multidrug Resistance, getestet. Durch die Kombination eines konventionellen Zytostatikums mit einem MDR-modulierenden Effluxpumpeninhibitor wird so die erforderliche, zelluläre Zytostatikakonzentration durch Hemmung des Transporters nicht mehr unterschritten. Die Proteinkinase Myt1 (PKMYT1) wurde als wichtiger Regulator des Cdk1/CyclinBKinase Systems biologisch charakterisiert. Die Myt1-katalysierte Phosphorylierung von Cdk1 bewirkt eine Inhibition des Proteins, die den Eintritt in die Mitosephase verhindert und somit den Zellzyklus reguliert. Eine Hemmung der Myt1-Kinase durch geeignete Inhibitoren führt somit zur vorzeitigen Aktivierung der Cdk1 und damit zur mitotischen Katastrophe - ein Wirkprinzip, was speziell für viele maligne Zelltypen zur Apoptose führt.
New potential inhibitors were synthesized by deduction of structure-activity-relationships of known MDR-modulators. They were tested by biochemical in vitro-investigations on different cancer cell lines concerning its inhibitory potential on P-glycoprotein, one of the most important efflux pumps in context with multidrug resistance. So the required cellular concentration can not fall under the limit by inhibition of the transport by combination of a conventional cytostatic drug with a MDR-modulating efflux pump inhibitor. Proteinkinase Myt1 (PKMYT1) was biologically characterized as important regulator of Cdk1/CyclinB kinase system. Myt1-cataliszed phosphorylation of Cdk1 effects an inhibition of the protein that prevents the entrance into mitose phase and regulation of the cell cycle. So inhibition of Myt1-kinase by a suitable inhibitor leads to premature activation of Cdk1 and to mitotic catastrophe - a mode of action that leads to apoptosis especially for malignant cell types.
