Abstract: Mit nur wenigen Ausnahmen werden alle messengerRNAs (mRNAs) kotranskriptional modifiziert. Eine dieser Modifikationen stellt die Spaltung und Polyadenylierung am 3‘-Ende der Prä-mRNAs dar. Der gebildete Poly(A)-Schwanz bestimmt durch Auswirkungen auf die Stabilität sowie die Effizienz von mRNA-Export und Translation maßgeblich das Schicksal einer jeder gebildeten mRNA. Obwohl der Prozess der Prä-mRNA-3‘-Prozessierung bereits seit über dreißig Jahren intensiv untersucht wird, ist es bislang nicht gelungen die Reaktionen von Spaltung und Polyadenylierung mit ausschließlich rekombinanten Proteinen in vitro ablaufen zu lassen.In dieser Arbeit wurde der zweite Reaktionsschritt der 3‘-Prozessierung, die Polyadenylierung, in vitro rekonstituiert und so die minimale Zusammensetzung an essentiellen Faktoren für die Katalyse dieses Schrittes der Prä-mRNA-3‘-Prozessierung identifiziert. Weiterhin wurden vielversprechende Vorversuche zur In-Vitro-Rekonstitution der Spaltungsreaktion durchgeführt.
Nearly all known messengerRNAs (mRNAs) are co-transcriptionally modified. One of these modifications is the cleavage and subsequent polyadenylation of the nascent mRNA at its 3’-end. The thus build poly(A) tail has significant influence upon the stability of the mRNA, as well as upon the efficiency of its export and translation. Thereby the poly(A) tail plays a decisive role in the fate of every produced mRNA. Despite of being investigated for over thirty years now, the reactions of cleavage and polyadenylation could yet not be reconstituted in an in vitro system with proteins from recombinant sources only. This work presents the reconstitution of the polyadenylation reaction from recombinantly produced proteins, defining the minimal set of essential protein factors for the catalysis of this reaction. Furthermore this work contains promising preliminary studies toward the in vitro reconstitution of the cleavage reaction.
