Abstract: Peroxisomenproliferator-aktivierte Rezeptoren (PPARs) sind kernständige Rezeptoren, die durch Bindung eines Liganden aktiviert werden und als Transkriptionsfaktoren die Genexpression regulieren. Als potente Liganden der PPARs wurden unter anderem Fettsäuren wie die konjugierten Linolsäuren (Abk. CLA) und oxidierte Fettsäuren identifiziert. Beide Gruppen von Fettsäuren sind biologisch hochaktive Substanzen, die im Organismus verschiedene physiologische aber auch pathophysiologische Wirkungen ausüben, ohne allerdings die zugrunde liegenden Ursachen zu verstehen. Aufgrund der Aktivierbarkeit von PPARs durch CLA und oxidierte Fettsäuren und der zentralen Rolle von PPARs in der Vermittlung von physiologischen und pathophysiologischen Wirkungen ist zu vermuten, dass diese Fettsäuren ihre Wirkungen über die Aktivierung von PPARs entfalten. Die in der vorliegenden Arbeit vorgestellten Untersuchungen beschäftigten sich deshalb schwerpunktmäßig mit der Aufklärung physiologischer und pathophysiologischer Wirkungen von CLA und oxidierten Fetten in zellulären Systemen und im Tiermodell unter besonderer Berücksichtigung von PPARs.
Peroxisome proliferator-activated receptors (PPARs) are nuclear receptors, which are activated by binding of a ligand and act as transcription factors regulating gene expression. Conjugated linoleic acids (CLA) and oxidized fatty acids have been identified as potent ligands of PPARs. Both groups of fatty acids are biologically active substances exerting various physiological but also pathophysiological actions in the organism, however, the underlying mechanisms of action are largely unknown. Because of the activation of PPARs by CLA and oxidized fatty acids and the central involvement of PPARs in both physiological but also pathophysiological actions it has been speculated, that the effects of these fatty acids are mediated by the activation of PPARs. The aim of the present investigations was to elucidate physiological and pathophysiological effects of CLA and oxidized fatty acids in cell systems and animal models taking PPARs into special consideration.
