Abstract: Darmkrebs ist einer der führenden Krebserkrankungen weltweit und betrifft in Deutschland jede 8. Krebserkrankung. Die Herunterregulierung der HLA-Klasse-I-Expression kann zu einer schlechten Prognose bei Krebspatienten beitragen. Es gibt nur begrenzte Informationen über die epigenetische und onkogenetische Regulation der HLA-Klasse-I-Molekülen, wobei mehrere Mechanismen beteiligt sein können. In der aktuellen Studie untersuchten wir die Korrelation zwischen dem MAPK-Signalweg und der HLA-Klasse-I-Expression. Resezierte kolorektale Tumorgewebe (n=54) wurden auf p-Erk- und HLA-Klasse-I-Expression mittels Immunhistochemie analysiert und bewertet. In dieser Arbeit konnte eine inverse Korrelation zwischen dem MAPK-Signalweg und der MHC-Molekül-Exprimierung im Kolorektalen Karzinom nachgewiesen werden. Der MAPK-Signalweg bietet eventuelle Angriffspunkte für zukünftige Therapien.
Colon cancer is one of the leading tumours in the world and it is considered among the big killers. Downregulation of HLA class I expression may contribute to a poor prognosis in cancer patients. There is limited information about epigenetic and oncogenic regulation of HLA class I, and multiple mechanisms may be involved. In the current study, we examined the relationship between the MAPK-signaling pathway and the expression of HLA class Resected colorectal tumor tissues (n = 54) were analyzed for p-Erk and HLA class I expression by immunohistochemistry. As a result there was a strong inverse correlation between p-Erk expression and HLA class I expression in clinical tumor samples. In conclusion, HLA-A expression is predominantly regulated by the MAPK pathway in colorectal cancer.
