Abstract: Das Pflanzenpathogen Xanthomonas campestris pv. vesicatoria (Xcv) verursacht die bakterielle Fleckenkrankheit auf Tomaten- und Paprikapflanzen. Ziel dieser Arbeit war die Identifikation neuer sRNAs aus Xcv und neuer RNA-Bindeproteine, welche eine Rolle in der Virulenz spielen. Es konnten 13 neue sRNAs identifiziert werden, deren Expression jedoch unabhängig von bekannten Virulenzregulatoren (HrpG und HrpX) ist. Pulldown- Experimente mit anschließender massenspektrometrischer Untersuchung identifizierten das sRNA-Bindeprotein DeaD, eine ATP-anhängige RNA-Helikase. Infektionsstudien von Xcv mit deaD-Mutanten zeigten eine deutlich verminderte Virulenz in Tomaten- und Paprikapflanzen, da die für die Pathogenität von Xcv essenziellen Regulatoren HrpG und HrpX durch DeaD reguliert werden. HrpG wird auf Transkript-Ebene und HrpX posttranskriptionell reguliert. Ein Wachstumseffekt einer deaD-Mutante im Vergleich zum Wildtyp wurde bei niedrigen Temperaturen nachgewiesen.; The plant pathogen Xanthomonas campestris pv. vesicatoria (Xcv) is the causal agent of bacterial spot disease and infects tomato and pepper plants. The aim of this work was to identify new sRNAs from Xcv and new RNA-binding proteins, which play a role in virulence. 13 new sRNAs could be identified of which the expression was independent of the main virulenve regulators, HrpG and HrpX. Pulldown experiments with following mass spectrometric analysis (MS-analysis) identified the sRNA binding protein DeaD, an ATP-dependent RNA helicase. Infection studies of Xcv with deaD mutants showed a significantly reduced virulence in tomato and pepper plants. Interestingly the regulators HrpG and HrpX, which are essential for the pathogenicity of Xcv, are regulated by DeaD. HrpG is regulated at the transcript level and HrpX posttranscriptionally. At temperatures below the optimum of Xcv it could be shown that the deaD mutant grew slower than the wild type.</description>

Abstract: Diese Studie zeigt, wie ein lokales Ammoniumangebot die Lateralwurzelverzweigung, durch Modulation des PIN-vermittelten polaren Auxintransports und der pH-abhängigen radialen Auxindiffusion, auslöst. Lokal appliziertes Ammonium unterdrückt die Expression von PIN2 und fördert die Expression von PIN4 in Lateralwurzelspitzen, was zu einer Auxinakkumulation im Zentralzylinder der Lateralwurzel in der Verzweigungszone führt. Zusätzlich fordert die Protonenfreisetzung, als Folge der AMT1;3-vermittelten Ammoniumaufnahme, die apoplastischen Ansäuerung. In Folge dessen, kommt es zu vermehrter Auxinprotonierung und verstärkter radialer Auxindiffusion, wodurch Auxin vom Zentralzylinder zu den äußeren Zellschichten der Wurzel gelangt. Hier löst Auxin das vermehrte Auswachsen von Lateralwurzel höherer Ordnung aus und stimuliert so die Bildung eines stark verzweigten Wurzelsystems. Die vorliegende Studie legt eine neue Grundlage, um zu verstehen, wie lokalisierte N-Verfügbarkeit die Auxinverteilung in Pflanzen moduliert und so die Lateralwurzelentwicklung neu programmiert, um die Wurzelarchitektur an sich verändernde Umweltbedingungen anzupassen.; This study explains how local ammonium triggers LR branching via modulating PIN-mediated polar auxin transport and pH-dependent radial auxin diffusion. Localized ammonium supply represses PIN2 expression and up-regulates PIN4 expression in LR tips, which promotes shoot-to-root auxin transport and leads to auxin accumulation in the vasculature of the LR branching zone. This process generates an auxin source for subsequent auxin diffusion. Meanwhile, proton release as a consequence of AMT1;3-mediated ammonium uptake leads to apoplastic acidification, auxin protonation and enhanced radial auxin diffusion, directing auxin from the vasculature to outer cell layers. The influence of local ammonium on auxin distribution synergistically stimulates LR emergence, shaping a highly branching root system. The present study lays a new basis to understand how localized N availability modulates auxin distribution in plants and subsequently reprograms LR development to alter root system architecture in a changing environment.</description>

Abstract: Pflanzliche, intrazelluläre NLR-Immunrezeptoren können Effektorproteine erkennen, die von pathogenen Mikroben in das Zytoplasma pflanzlicher Zellen sekretiert werden. Eine Hauptklasse der NLRs besitzt eine N-terminale TIR-Domäne (TNLs). Für eine Immunaktivierung durch TNLs sind EDS1-basierende Heterokomplexe essentiell. Die präzise Rolle der EDS1-Heterokomplexe in der Immuninduktion und/oder Signalweiterleitung ist bislang nicht verstanden. In dieser Arbeit wurde Nicotiana benthamiana (Nb; Solanaceae) als neuer Modellorganismus etabliert, um die Funktionen des Lipase-ähnlichen Proteins EDS1 zu analysieren und um die pflanzlichen Immunsignalwege zwischen Arabidopsis thaliana (At; Brassicaceae) und Nb zu verglichen. EDS1-basierete Heterokomplexe benötigen zwei Proteinen unterschiedlicher phylogenetischer Herkunft, für eine Immunreaktion PAD4 in At bzw.SAG101 in Nb. Dies deutet auf eine Koevolution von EDS1-Komplexen mit zusätzlichen Wirtsfaktoren hin, wahrscheinlich Proteininteraktoren. Ferner gibt es Hinweise auf eine vorherrschende Rolle des EDS1-SAG101 Komplexes in der pflanzlichen Immunität, wie in Nb beobachtet.; Plant intracellular immune receptors of the NLR-type can detect effector proteins, which are secreted into the plant cell cytoplasm during the infection process by pathogenic microbes. One major subclass of NLRs, termed TNLs, contain an N-terminal TIR domain. The induction of immune responses by TNLs critically depends on presence of heterodimeric complexes based on the lipase-like protein EDS1. However, the precise role of EDS1 complexes in immune induction and/or signal transduction remains unknown. In this work, Nicotiana benthamiana (Nb; Solanaceae) was established as a new model system for the analysis of EDS1 functions, and immune signaling pathways were compared between Arabidopsis thaliana (At; Brassicaceae) and Nb. Intriguingly, heterocomplexes of EDS1 with either PAD4 or SAG101, two EDS1-interacting proteins of distinct phylogenetic origins, function in immune signaling in At or Nb, respectively. This suggests co-evolution of EDS1 complexes with additional host factors, most likely protein interactors. Furthermore, evidence is provided for a prevalent role of EDS1-SAG101 in immune signaling, as observed in Nb.</description>

Abstract: In den letzten Jahren wurde festgestellt, dass eukaryotische nicht-kodierende RNAs eine wichtige Rolle in einer Vielzahl von Prozessen spielen, die von der Regulation der Genexpression bis zur Translation reichen. Bei Pflanzen hat sich gezeigt, dass sie an einer Vielzahl biologischer Prozesse beteiligt sind, darunter Blütezeitregulation, Wurzelentwicklung sowie Hormon- und Stressreaktionen. Natürliche Antisense-lange nicht-kodierende RNAs (lncNATs), ein Subtyp von lncRNAs, die vom komplementären DNA-Strang eines Protein-kodierenden Gens transkribiert werden, haben die Entwicklung verschiedener Pflanzenarten beeinflusst. Anhand des Modellorganismus Arabidopsis thaliana untersuchten wir das regulatorische Potential einer lncNAT, die das Gen UDP-GLYCOSYLTRANSFERASE 76E12 (UGT76E12) überlappt.; In recent years, eukaryotic non-coding RNAs were found to play an important role in a variety of processes ranging from gene expression regulation to translation. In plants, they have been shown to be involved in a wide range of biological processes including flowering time regulation, root development, and hormone and stress responses. Natural antisense long non-coding RNAs (lncNATs), a sub-type of lncRNAs that are transcribed from the complementary DNA strand of a protein-coding gene, have been revealed to influence development in various plant species. Using the model organism Arabidopsis thaliana, we investigated the regulatory potential of a lncNAT which overlaps the UDP-GLYCOSYLTRANSFERASE 76E12 gene (UGT76E12).</description>
