dc.description.abstractEs wurden Untersuchungen zum Stoffübertritt im Xylem und im Phloem zwischen dem Holoparasiten Cuscuta spec. und seinen Wirtspflanzen durchgeführt. Neben Experimenten zur Translokation von Saccharose, Aminosäuren, Xenobiotika und Fluoreszenzfarbstoffen erfolgten auch Versuche zum Transport von Viren und Makromolekülen (GFP). Die offenen funktionellen Verbindungen zwischen den Leitbündeln des Wirtes und des Parasiten wurden durch die Fluoreszenzfarbstoffe Texas red Dextran (Xylem) bzw. Carboxyfluorescein und Hydroxypyrentrisulfonsäure (Phloem) dargestellt. Der Transport xylemmobiler (Atrazin, DCMU, Nicotin, Cadmium) Substanzen aus dem Wirt in den Parasiten ist sehr gering und nicht selektiv. Ambimobile (Glyphosat, AIB) Verbindungen wurden in wesentlich höheren Anteilen im Parasiten gemessen. Diese Substanzen gelangen nach einem Xylem-Phloem-Transfer in den Internodien des Wirtes hauptsächlich über das Phloem in die Cuscuta. Der Transport über das Phloem ist wesentlich intensiver als über das Xylem und ebenfalls nicht selektiv. Für Saccharose, den Virus PVY und für das GFP wurde in Brückenversuchen neben einem Transport aus dem Wirt in den Parasiten auch ein Transport aus dem Parasiten in den Wirt nachgewiesen. Die Ergebnisse der Untersuchungen zur Translokation von Assimilaten, Xenobiotika, Fluoreszenzfarbstoffen, insbesondere aber der Transfer des Virus und des GFP, zwischen Cuscuta und ihren Wirtspflanzen beweisen eindeutig einen symplastischen Phloemkontakt. Für die Allgemeingültigkeit dieser Aussage sprechen übereinstimmende Ergebnisse zum CFTransport mit Wirtspflanzen aus vier unterschiedlichen Familien und Cuscuta reflexa, sowie der erfolgreiche GFP-Transport aus transgenen Arabidopsis-Pflanzen in drei unterschiedliche Cuscuta-Arten.-
dc.description.abstractThe transport of substances in the xylem and the phloem from compatible host plants into the parasite Cuscuta spec. was investigated. For translocation experiments not only sucrose, amino acids, xenobiotics and fluorescent dyes but also the virus PVY and GFP were used. The uninterrupted functional connections between host plants and Cuscuta were demonstrated by a continuous fluorescence labeling by texas red (xylem), CF or HPTS (phloem), respectively. The transport of xylem mobile substances from host to the parasite shows a very low extent and is non selective. Ambimobile compounds were found at a higher level within the parasite. They could transported via the phloem into the sink tissue of Cuscuta. That requires a xylemphloem- transfer of the ambimobile compounds after their uptake by host roots. The transfer takes place within internodes. The phloem transport is more intensive and also non selective. Experiments with Cuscuta-"bridges" connecting two hosts have shown a transport of sucrose, virus and GFP not only from host to parasite but also from the parasite into a second host plant. The results of the translocation experiments with assimilates, xenobiotics, fluorescent dyes, especially the transfer of viruses and GFP, indicate undoubtedly a symplastic phloem connection between Cuscuta and its host plants. The general validity of this statement is given by the corresponding results of CF-transport between Cuscuta reflexa and host plants belonging to 4 different families as well as the successful GFP-transport between transgenic Arabidopsis plants and 3 different Cuscutaspecies.eng
