dc.description.abstractCalystegine sind polyhydroxylierte Nortropanalkaloide, die sich in ihrer Biosynthese von der Tropan-alkaloidbiosynthese ableiten. Calystegin A3 und B2 akkumulieren als Hauptalkaloide in der Convolvulaceae Calystegia sepium in allen Pflanzenorganen. Es wurden zwei cDNA-Sequenzen mit hoher Homologie zu Tropinonreduktasen (TR) isoliert. Beide Sequenzen zeigen die typischen Merkmale der kurzkettigen Dehydrogenasen/Reduktasen. Eine cDNA-Sequenz wurde in E. coli heterolog exprimiert. Dieses Enzym reduziert Tropinon ausschließlich zu Tropin und stellt damit eine TRI dar. Die Expressionsanalyse der trI in den einzelnen Pflanzenorganen ergab eine hohe Transkriptionsrate in der Pflanzenwurzel und ein schwaches Signal in der Sprossachse. Die Untersuchung der genomischen Struktur der trI im Southern Blot lässt auf eine Genfamilie mit mindestens 3 verschiedenen Genen schließen. Die zweite Sequenz konnte heterolog weder in Bakterien, noch in Hefe oder einem Pflanzensystem exprimiert werden und wird als putative TR betrachtet. Das native Protein der pseudotropinbildenden Tropinonreduktase (TRII) wurde aus Wurzelkulturen isoliert, angereinigt und charakterisiert. Es ähnelt in seinen biochemischen Eigenschaften den bisher bekannten TRII der Solanaceen. Der Verlauf der Biosynthese vom Tropinon über Pseudotropin zu den Calysteginen wurde mit einer 15N-Tropinonfütterung belegt. Der Einbau der Markierung wurde sowohl mit NMR als auch quantitativ mit GC-MS analysiert. Norpseudotropin konnte als weiterer Metabolit der Calysteginbiosynthese identifiziert werden.-
dc.description.abstractCalystegines are polyhydroxylated nortropanes which derive from the tropane alkaloid biosynthesis. Calystegine A3 and B2 accumulate as main alkaloids in the Convolvulaceae Calystegia sepium in all plant organs. Two cDNA sequences with high homologies to known tropinone reductases (TR) were isolated. Both sequences show typical motifs of the short chain dehydrogenase/reductase family. One cDNA was heterolog expressed in E.coli. Tropinone reduction by the enzyme led to formation of tropine exclusively, justifying the designation TRI. Transcript abundance of trI in all plant organs revealed a high transcription rate in root tissue, and a weak expression was seen in stem tissue. Investigation of the genomic structure of trI by Southern blotting indicates a gen family with minimum 3 different genes. The second sequence could be expressed neather in bacteria, nor in yeast or plants and is therefor designated as a putative TR. The native protein of the pseudotropine forming tropinone reductase (TRII) was isolated from root cultures, purified and characterised. It is similar in its biochemical properties to known TRII of the Solanaceae family. By application of 15N-labeled tropinone the biosynthesis of calystegines from tropinone over pseudotropine was prooved. The incorporation of label was analysed by NMR and quantified by GC-MS. Norpseudotropine was identified as a further metabolit of the calystegine biosynthesis.eng
