dc.description.abstractImmunotoxine sind chimäre Proteine, bestehend aus einer zellbindenden und cytotoxischen Komponente. Sie werden hauptsächlich bei der Behandlung von Krebs eingesetzt. In der vorliegenden Arbeit wurden drei modulare Immunotoxine konstruiert und charakterisiert, bei denen die zellbindende Komponente und die cytotoxische Komponente mittels polyionischer Fusionspeptide verknüpft sind. Mit diesem Prinzip konnte ein Kopplungssystem etabliert werden, das in Zukunft ein schnelles und effizientes screening der für eine bestimmte Therapie geeigneten Kombination aus Antikörperfragment und Toxin gestattet. Als zellbindende Komponente wurde das disulfid-verbrückte B3-Antikörperfragment verwendet. Es wurden 3 verschiedene cytotoxische Proteine eingesetzt, eine verkürzte Variante des Pseudomonas Exotoxins (PE38), Granzym B (GzmB) und eine modifizierte β-Galaktosidase (toxGal). Am Beispiel der beiden polyionischen Immunotoxine B3-PE38 und B3-GzmB konnte gezeigt werden, dass weder die polyionische Fusion noch die Kopplung der beiden Komponenten einen Einfluß auf deren Stabilität und biologische Aktivität haben. Entscheidend jedoch für den Einsatz von Immunotoxinen in der Therapie ist ihre Zelltyp-spezifische und cytotoxische Wirksamkeit. Für B3-PE38 und B3-GzmB konnte dies in Zellkulturexperimenten nachgewiesen werden. Im Gegensatz zu den beiden ersten Immunotoxinen konnte für B3-toxGal, das aus dem B3-Antikörperfragment und einer mit fluorierten Tyrosinen modifizierten Variante der β-Galaktosidase aufgebaut ist, keine signifikante toxische Wirkung auf Zellen beobachtet werden.-
dc.description.abstractImmunotoxins are chimeric proteins consisting of a cell binding and cytotoxic component. They are primarily used for cancer therapy. In the present study three different modular immunotoxins have been constructed and characterized. In all three cases the cell binding and cytotoxic component are connected via polyionic fusion peptides. Thus, a new coupling system has been established, enabling a fast and efficient screening for an optimized combination of cell binding and cytotoxic component, suitable to apply in certain anticancer therapies. As a cell binding target module the disulfide bridged B3 antibody fragment was used. Different cytotoxic components have been used, a truncated variant of the Pseudomonas exotoxin (PE38), granzyme B (GzmB) and a modified β-galaktosidase (toxGal). The biophysical characteristics of the polyionic immunotoxins B3-PE38 and B3-GzmB have been investigated. Neither the polyionic fusion peptides nor the coupling of the components have influenced their stability or biological activity. An essential requirement of immunotoxins is their cell type specific targeting and cytotoxic activity. This has been demonstrated in cell culture experiments for the immunotoxins B3-PE38 and B3-GzmzB. The third polyionic immunotoxin B3-toxGal consists of the B3 antibody fragment and a modified variant of β-galaktosidase with fluorinated tyrosines, which are toxic for cells. However, no significant cytotoxic effect could be shown for this immunotoxin.eng
