Abstract: Biomedical high-entropy alloys (bio-HEAs) represent a promising class of materials for biomedical applications due to their combination of excellent biocompatibility, corrosion resistance, and distinctive mechanical properties. However, owing to their typically complex compositions it is difficult to assign specific properties to specific elemental components – and thus to make comprehensible adjustments. The aim of this work is to address this challenge by examining a simplified ternary alloy system composed of the biocompatible elements Ta, Nb, and Ti – referred to as TNT-type alloys – across defined compositions and establish a fundamental understanding of the system as a basis for future alloying strategies. The compositions Ta-Nb-xTi (x = 10, 33.3, 50, 70 at. %) were chosen for the examinations to not only analyse the equimolar composition per se, but also to investigate the influence of titanium variations on the alloy properties. Preliminary thermodynamic calculations based on the binary compositions, the principles of HEAs, as well as Ti-alloys were all in good agreement and predicted the theoretical formation of a single-phase microstructure for all compositions. Subsequently, the alloys were synthesised by arc melting, which resulted in pronounced dendritic microstructures. Through a following heat treatment study, the parameters for a complete homogenisation of all compositions and thus single-phase body-centred cubic (bcc) microstructure were obtained. The microstructural evolution was characterised and validated using optical microscopy, scanning electron microscopy (SEM) and X-ray diffraction analysis (XRD) techniques. The characterisation of the samples surfaces by means of confocal microscopy after metallographic preparation yielded comparable (smooth) surfaces to reference samples of pure Ta, Nb and Ti, as well as alloy Ti-6Al-4V (TAV), which was a favourable starting point for the subsequent biocompatibility trials. Moreover, the mechanical properties of the TNT-type alloys were evaluated through hardness testing, room-temperature compression, and 3-point bending on microsamples and compared to the reference alloy TAV. While all alloys showed pronounced plastic deformability, the results indicated a strong correlation between the mechanical characteristics, the Ti-content and the homogenisation conditions. Furthermore, all alloys exhibited greater elastic deformability and lower flexural moduli than the reference alloy, with the equimolar Ta-Nb-Ti composition showing the most balanced combination of hardness, flexural strength and compressive properties. In addition, a biocompatibility assessment consisting of an MM6 inflammatory reaction test and an osteoblast (SaOs-2) attachment and subsequent mineralisation assay was performed, which showed comparable results to the established TAV reference thus indicating no negative influence of the TNT-type alloys on the overall biocompatibility. Furthermore, the antibacterial capacity of the alloys was assessed via E. coli and S. capitis bacteria in vitro, generally demonstrating composition-dependent anti-adhesive properties against both types of bacteria. In a next step, the S. capitis trials were transferred to a G.mellonella larvae model in vivo and the previous results could be confirmed by reduced mortality rate of larvae compared to the reference TAV. Finally, basic electrochemical investigations by means of potentiostatic measurements were performed in order to gain an initial impression of the corrosion behaviour of the TNT-type alloy compositions examined and thus be able to draw conclusions about their biocompatible and anti-adhesive performance.
Biomedizinische Hochentropielegierungen (Bio-HEAs) sind aufgrund ihrer exzellenten Biokompatibilität, Korrosionsbeständigkeit und mechanischen Eigenschaften eine attraktive Materialklasse für biomedizinische Anwendungen. Die meist komplexen Legierungszusammensetzungen dieser Werkstoffe erschweren jedoch die eindeutige Zuordnung spezifischer Eigenschaften zu einzelnen Legierungselementen und damit eine gezielte Legierungsanpassung. Das Ziel der vorliegenden Arbeit ist es, an dieser Stelle anzusetzen und ein vereinfachtes ternäres Legierungssystem aus den biokompatiblen Elementen Ta, Nb und Ti (TNT-type Alloys) in definierten Zusammensetzungen zu untersuchen, um ein grundlegendes Verständnis des Legierungssystems zu schaffen, bevor es um weitere Elemente erweitert wird. Die Zusammensetzungen Ta-Nb-xTi (x = 10, 33,3, 50, 70 at. %) wurden gewählt, um nicht nur die äquimolare Zusammensetzung zu analysieren, sondern auch den Einfluss von Ti-Variationen auf die Legierungseigenschaften zu untersuchen. Thermodynamische Berechnungen der Legierungen – basierend auf den binären Systemen, HEA-Prinzipien und Ti-Legierungen – stimmten gut überein und prognostizierten für alle Zusammensetzungen die Bildung einer einphasigen Mikrostruktur. Im Folgenden wurde dies experimentell verifiziert durch eine Synthetisierung der Legierungen mittels Lichtbogenschmelzverfahren. Die ausgeprägten dendritischen Mikrostrukturen nach dem Schmelzprozess konnten durch eine anschließende Wärmebehandlungsstudie erfolgreich in einen vollständig homogenisierten, kubisch-raumzentrierten (krz) Mischkristall überführt werden. Die mikrostrukturelle Entwicklung wurde mittels Lichtmikroskopie, Rasterelektronenmikroskopie (REM) und Röntgendiffraktometrie (XRD) charakterisiert und validiert. Die Charakterisierung der TNT-type alloy Probenoberflächen mittels Konfokalmikroskopie nach metallographischer Präparation zeigte vergleichbar glatte Oberflächen zu Referenzproben aus reinem Ta, Nb, Ti sowie der Referenzlegierung Ti-6Al-4V (TAV), was einen günstigen Ausgangspunkt für nachfolgende Biokompatibilitätstests darstellte. Die mechanischen Eigenschaften der TNT-type Alloys wurden durch Härteprüfung, Raumtemperatur-Druckversuche und 3-Punkt-Biegung an Mikroproben evaluiert und mit TAV verglichen. Grundsätzlich zeigten alle Legierungen ausgeprägte plastische Verformbarkeit, wobei die Ergebnisse auf eine starke Korrelation zwischen den mechanischen Kennwerten, dem Ti-gehalt sowie dem Wärmebehandlungszustand schließen ließen. Zudem zeigten alle Legierungen höhere elastische Verformbarkeit und niedrigere Biegemoduln als TAV auf, wobei die equimolare Ta-Nb-Ti-Zusammensetzung die ausgewogenste Kombination von Härte, Biegefestigkeit und Druckeigenschaften aufwies. Die Biokompatibilität der Legierungen wurde mittels MM6-Entzündungsassay und Osteoblasten (SaOs-2) Besiedlungs- und Mineralisationsversuchen untersucht, welche vergleichbare Ergebnisse zur etablierten Referenz TAV lieferten und somit keinen Hinweis auf einen negativen Effekt der TNT-type Alloys geben. Die antibakterielle Kapazität der Legierungen wurde mittels E. coli und S. capitis in vitro untersucht und zeigte generell zusammensetzungsabhängige anti-adhäsive Eigenschaften gegen beide Bakterientypen. Die S. capitis-Versuche wurden auf ein G. mellonella-Larvenmodell in vivo übertragen, wobei die Ergebnisse durch reduzierte Mortalitätsraten im Vergleich zu TAV bestätigt wurden. Abschließend wurden grundlegende elektrochemische Untersuchungen mittels potentiostatischer Messungen durchgeführt, um einen ersten Eindruck vom Korrosionsverhalten der untersuchten Legierungszusammensetzungen zu gewinnen und so Rückschlüsse auf ihre biokompatible und anti-adhäsive Performance ziehen zu können.
