Abstract: Die Metallopeptidasen APN, NEP und DPPIV besitzen als Bestandteile von Membranmikrodomänen auch eine enzymunabhängige Funktion. Mittels durchflusszytometrischer FRET-Messung zeigten wir eine Kolokoalisation zwischen APN und Fc-Rezeptoren-I und -II auf U937- und THP-1-Zellen, zwischen APN und HLA-DR auf U937-Zellen und zwischen APN und CD11b/CD18 auf THP-1-Zellen. Die Bestätigung der Klolokalisation zwischen APN und CD32 erfolgte mittels Koimmunopräzipitation auf THP-1-Zellen. Auf U937-Zellen führte die Stimulation mit TPA zur signifikanten Steigerung der Expression von CD13 und CD64, aber nicht von HLA-DR. Der Abstand zwischen CD13 und HLA DR verringerte sich, während der Abstand zwischen Fc-Rezeptor-II und-I und APN gleich geblieben ist. Auf THP-1-Zellen führte die Stimulation mit IL-4 zur Steigerung der Expression von APN, Wegrücken der CD13 von CD64 und Hinrücken der CD13 zu CD32. FRET-Messung mit FACS ist geeignet zur Untersuchung der Beziehungen auf der Zelloberfläche.
The metallopeptidases APN, NEP and DPPIV have enzyme-independent functions in the membrane and are constituents of membrane microdomains. Using flow cytometric FRET measurements, we were able to demonstrate a colocalization between APN and Fc receptors-I and -II on U937-and THP-1-cells, between APN and HLA-DR on U937 cells and between APN and CD11b/CD18 on THP-1-cells. The relationship between APN and CD32 was able to be confirmed with co-immunoprecipitation on THP-1-cells. Th e stimulation with TPA on U937 cells, led to a significant increase in expression of CD13 and CD64, but not of HLA-DR. e distance between CD13 and HLA DR decreased, while the distance between Fc receptor II and -I and APN remained the same. Th e stimulation with IL-4 on THP-1-cells, led to increase in expression of APN, moving away the CD13 from CD64 and moving the CD13 to CD32. FRET measurement with FACS is suitable for examination of the relationships on cell surface.
