Abstract: In der vorgelegten Dissertation wurde der Einfluss der katalytischen β-Untereinheit von Calcineurin (PPP3CB) hinsichtlich ihrer Signalwege und Funktion in Gefäßen, anhand eines transgenen PPP3CB Knockout (KO) Mausmodells, untersucht. Unter basalen Bedingungen waren keine Unterschiede zwischen Wildtyp (WT) und KO Mäusen detektierbar. Ang II als Mediator für kardiale Hypertrophie und strukturelles Remodeling, wurde männlichen KO Mäusen chronisch über osmotische Minipumpen verabreicht. Der systolische Blutdruck (SB) wurde über die tail cuff Methode gemessen und zeigte keine Ang II-induzierte Erhöhung des SB in PP3CB KO Mäusen im Vergleich zu WT Tieren. Histologische Färbungen von der Aorten männlicher KO Mäuse zeigten keine signifikant erhöhte Mediadicke im Vergleich zu WT Tieren. Die Transkriptom Analyse der Aorten Ang II-behandelter Mäuse ergab eine erhöhte Expression von Fibrosemarkern in WT Tieren, nicht aber in KO Mäusen. Die Deletion von PPP3CB erwies sich als förderlich.
The submitted dissertation investigates the influence of the catalytic beta subunit of calcineurin (PPP3CB) with regard to signaling pathways and function in vessels by means of a transgenic PPP3CB knockout (KO) mouse model. Under basal conditions no differences in the genotypes were detectable. AngII as mediator of cardiac hypertrophy and structural remodeling was applied chronically to adult male KO and Wildtype (WT) mice by osmotic minipumps. Systolic blood pressure (SBP) was measured via tailcuff method and revealed angII-mediated SBP increase was attenuated in PPP3CB KO mice but not in WT mice. Histological stainings of aortic sections showed no significantly increased media thickness in male angII-treated KO mice compared to WT animals. Transcriptome sequencing of aortic samples followed by gene enrichment analysis revealed an increased expression of fibrosis markers in angII-treated WT animals but not in KO mice. The deletion of PPP3CB turned out to be beneficial.
