Abstract: Die vorliegende Arbeit beschäftigt sich mit der thermodynamischen und kinetischen Beschreibung von Entfaltungsvorgängen monoklonaler Antikörper (mAb) zu deren Stabilitätsvorhersage. Daher wurden Parameter wie Schmelztemperaturen (Tm), van´t Hoff Enthalpien (∆Hvh) und Aktivierungsenergien (Ea) bestimmt, um ihre prädiktive Aussagekraft hinsichtlich thermischer Antikörperstabilitäten zu evaluieren. Dazu wurden temperaturabhängige intrinsische Fluoreszenzmessungen (IF) aufgenommen und aus den jeweiligen Entfaltungsprofilen thermodynamische und kinetische Parameter mittels zweier verschiedener scheinbarer Drei-Zustands-Modelle bestimmt. Als bester prädiktiver Parameter hinsichtlich thermischer Stabilität von Antikörpern wurde in dieser Studie die Aktivierungsenergie der CH2-Domäne gefunden. Zusätzlich wurden in dieser Arbeit zwei neue Methoden entwickelt, um Entfaltungsübergänge den jeweiligen mAb-Domänen zuordnen zu können, auch ohne den Vergleich zu kalorimetrischen Messungen.
The present study deals with the analysis of thermodynamics and kinetics of monoclonal antibody (mAb) unfolding in solution to estimate their thermal stability. Therefore, parameters like melting temperatures (Tm), van´t Hoff enthalpies (∆Hvh), and activation energies (Ea) were tested to correlate with thermal stability to investigate their predictive character. Temperature dependent intrinsic fluorescence (IF) thermograms were recorded and values of ∆Hvh and Ea were determined by applying appropriate fit functions based on two different apparent three-state models. For thermal stability prediction, Ea of the CH2-domain was investigated to show strongest correlation with thermal stability. Additionally, two novel methods were developed and tested to assign transitions directly to mAb-domains from IF thermograms without the comparison to differential scanning calorimetry (DSC).
