Abstract: TAL- (transcription activator-like) Effektoren sind Virulenzfaktoren der phytopathologen Bakterien der Gattung Xanthomonas. Sie werden während der Infektion in die Wirtzelle eingeschleust und verändern dort die Transkription der pflanzlichen Wirtszelle. Dabei fungieren diese Proteine als Transkriptionsfaktoren. Die TAL-Effektoren besitzen eukaryotische Kernlokalisierungssignale, für den Zellkernimport und eine saure Aktivierungsdomäne, die jeweils essentiell für die Funktion sind. Im Rahmen dieser Arbeit konnte gezeigt werden, dass die DNA-Bindung und die Spezifität der TAL-Effektoren durch die in der zentralen Region des Proteins befindlichen 34-Aminosäure umfassenden Sequenzwiederholungen (sogenannte repeats) bestimmt wird. Dabei wurde deutlich, dass ein repeat zu einem Basenpaar korreliert. Die DNA-Bindung an vorhergesagte Zielsequenzen konnte mit natürlichen und künstlich generierten TAL-Effektoren in planta und in humanen Zellen gezeigt werden.
TAL (transcription activator-like) effectors are virulence factors of phytopathogenic bacteria of the genus Xanthomonas. They are translocated during the infection into the host cell and alter the transcription of the plant host cell. In this case, these proteins function as transcription factors. The TAL effectors have eukaryotic nuclear localization signals, for nuclear import in to the cell and an acidic transcriptional activation domain, which are each essential for the function. In this thesis it was shown that the DNA-binding and specificity of TAL effectors is determined by the central region of the protein containing 34-amino acid sequence repeats. It became clear that one repeat correlate to a base pair. The DNA-binding to predicted target sequences could be shown with natural and artificially generated TAL effectors in planta and in human cells.
