Abstract: Es wurde gezeigt, dass die Agrobacterium tumefaciens-vermittelte Transformation (ATMT) zur Identifizierung von Virulenzgenen des Maispathogens Colletotrichum graminicola effizient einsetzbar ist. Aus einer Transformantenbank wurden 19 virulenzreduzierte oder apathogene Mutanten ermittelt. Nach Identifizierung von T-DNA Integrationsstellen, konnten die Gene für einen V-ATPase Assemblyfaktor, eine Allantoicase und ein hypothetisches Protein als putative Virulenzgene von C. graminicola ermittelt werden. Mit einer Gendeletionsanalyse wurde das Gen CgAAF1, das für einen ER-membranständigenV-ATPase Assemblyfaktor codiert, detailliert als Virulenzgen charakterisiert. Die Penetration des Pflanzengewebes war reduziert und eine erhöhte pflanzliche Abwehr in Form verstärkter Papillenbildung wurde beobachtet. Eine mögliche Beeinträchtigung von Sekretionsvorgängen wird als Ursache für die reduzierte Virulenz der Deletionsmutante diskutiert.
It has been shown that Agrobacterium tumefaciens-mediated transformation (ATMT) is suitable to identify virulence genes of the maize pathogen Colletotrichum graminicola. 19 mutants with a reduced virulence or with a loss of pathogenicity have been found in a transformant library. After the identification of T-DNA integration sites, the genes for a VATPase assembly factor, for an allantoicase and for a hypothetical protein have been regarded as virulence genes of C. graminicola. Gene deletion analyses characterized in detail the virulence gene CgAAF1, encoding an V-ATPase assembly factor localized in the ER membrane. The colonialization of the plant by the deletion strain was dramatically reduced, reduced penetration of the plant tissue and an increased plant defence by increased papillae formation was observed. An impairment of fungal secretion processes are discussed as the reason for the reduced virulence of the deletion strain.
