Abstract: Trockenzeiten zählen heute zu den Hauptgründen für Ernteausfälle und damit verbundene Hungersnöte. Ein Ansatz zur Lösung dieses Problems ist die Applikation von Pflanzenwachstumsregulatoren, die die Trockenheitstoleranz steigern. Auf der Suche nach solchen Substanzen ist es wichtig, robuste Assaysysteme zu entwickeln, die eine Hochdurchsatzanalyse ermöglichen. Daher wurde ein in vivo Assay auf Basis von Lemna minor entwickelt. Durch Biomarkeranalysen konnte bestätigt werden, dass die Ergebnisse auf Kulturpflanzen übertragbar sind. Um Anhaltspunkte zum Wirkmechanismus von aktiven Verbindungen zu erhalten, wurde eine Screeningroute, basierend auf Inhibitoren von PARP-Enzymen etabliert. Mittels der etablierten Screeningroute bestehend aus Protein Assay, Hefe Assay sowie Lemna Assay wurden Substanzdatenbanken untersucht. Insgesamt konnten in dieser Arbeit 55 neue AtPARP-1-Inhibitoren und 10 Substanzen identifiziert werden, die die Trockenstresstoleranz von L. minor steigern.
Drought spells are one of the main reasons for today’s yield losses in agriculture. One approach to solve this problem is the application of plant growth regulators that increase plant’s stress tolerance. In this course robust assays are required, which allow high-throughput analysis of compound libraries. Therefore a fast, whole-plant assay using Lemna minor was developed. The ability to transfer obtained results to crop plants was shown using biomarker analysis. To gain insight into the mode of action of identified compounds, a screening route was established focusing on inhibitors of PARP1. Using the screening route composing of a protein assay, a yeast assay and the in vivo Lemna assay, a compound library of 121 substances was screened. In this course 52 new AtPARP-1 inhibitors were identified of which 7 compounds revealed a drought stress tolerance inducing activity.
