Abstract: Mutationen oder die Abwesenheit vieler Proteine wie auch der desmosomalen Proteine Plakoglobin und Plakophilin und des Z-Scheiben-Proteins ZASP/Cypher stehen mit Herzerkrankungen wie ARVC und DCM in Verbindung. In dieser Studie sollte der Frage nachgegangen werden, ob die Expression der Proteine Plakoglobin, Plakophilin und ZASP/Cypher einer altersabhängigen Expression unterliegt. Dafür wurden 53 Proben männlicher Patienten (Durchschnittsalter 67) mittels Western Blotuntersucht. Alle Patienten waren mit β-Blockern behandelt worden, die meisten auch mit ACE-Hemmern, ASS und Statinen. Das mittlere NYHA-Stadium lag bei 2. Die Ergebnisse der Studie sind nicht eindeutig. Eine lineare Korrelation konnte nicht nachgewiesen werden, wohl aber signifikante Unterschiede zwischen Altersgruppen. Es konnten außerdem eine positive Korrelation zwischen ZASP/Cypher und Plakoglobin und signifikante Einflüsse von ASS und ACE-Hemmern auf die Plakoglobin-Expression festgestellt werden
Mutations or the absence of proteins like desmosomal proteins Plakoglobin and Plakophilin and of z-disc proteins like ZASP/Cypher are associated with heart diseases like ARVC and DCM. In this study the question should be answered if the expression of Plakoglobin, Plakophilin and ZASP/Cypher is underlying an age-dependency. Western blots of 53 male patients (average age 67) were done. All patients were treated with beta-blockers, most of them as well with ACE inhibitors, ASS and statins. Average NYHA stadium was 2. Results of this study were not clear. A linear correlation could not be found even though there are significant differences between age groups. Furthermore there is a positive correlation between expression of ZASP/Cypher and Plakoglobin and significant effects of ASS and ACE-inhibitors on plakoglobin expression could be found.
