dc.description.abstractIn dieser Arbeit wurden Synovial- und Leiomyosarkome immunhistochemisch untersucht. Wir wollten wissen, welche prognostische Bedeutung Survivin in diesen beiden Weichteilsarkomentitäten hat. Survivin ist ein Mitglied der Inhibitoren der Apoptose und ist zudem eng mit der Zellteilung verknüpft. Survivin ist in vielen Malignomen überexprimiert. Außerdem wollten wir wissen ob die beiden Hypoxiemarker Carboanhydrase9 und Osteopontin ebenfalls eine prognostische Relevanz in beiden Entitäten aufweisen. Dafür stand ein Tumorkollektiv von 26 Synovial- und 24 Leiomyosarkomen zur Verfügung. Die Färbungen wurden für die entsprechenden Proteine Survivin, Carboanhydrase9 und Osteopontin durch die Avidin-Biotin-Methode angefertigt und mittels immunoreaktiven Score nach Remmele und Stegner ausgewertet. Die statistische Auswertung erfolgte durch die multivariate Cox-Regressionsanalyse. In der vorliegenden Arbeit ist es gelungen, Survivin als unabhängigen prognostischen Faktor in Synovial- und Leiomyosarkomen zu identifizieren. Weiterhin konnte eine signifikante Assoziation zwischen der Proteinexpression für CA9 und OPN und dem tumor-assoziierten Überleben für Synovialsarkome aufgezeigt werden.-
dc.description.abstractIn this study, synovial and leiomyosarcomas were examined by immunohistochemistry. We were interested if survivin detected by immunohistochemistry has prognostic relevance especially for patients with the two soft tissue sarcoma entities. Survivin, a member of the inhibitor of apoptosis-protein family suppresses apoptosis and regulates cell division. It is strongly overexpressed in the vast majority of cancers. We were also interested if the hypoxia markers carboanhydrase9 and osteopontin detected by immunohistochemistry has prognostic relevance in both entities. Tumors of leiomyosarcoma (n = 24) and synovial sarcoma patients (n = 26) were investigated for their expression of survivin, carboanhydrase9 and osteopontin by immunohistochemistry. The expression of theproteins was assessed in tumor cells using an immunoreactive scoring system (IRS). The Statistical analysis was performed by multivariate Cox-regression analysis. Survivin protein expression in the cytoplasma and in the nucleus detected by immunohistochemistry is significantly associated with prognosis of leiomyosarcoma and synovial sarcoma patients. Furthermore, a significant association between the protein expression of CA9 and OPN and the tumor-associated survival for Synovialsarkome, could be shown.eng
