dc.description.abstractDer Akkumulation von altersabhängigen Proteinmodifikationen durch Kohlenhydrate (Advanced glycation endproducts, AGEs) wird eine wichtige Rolle bei der Ätiologie kardiovaskulärer Erkrankungen zugeschrieben. Altern des humanen Myokards ist charakterisiert durch strukturelle und funktionelle Veränderungen, die Einfluss auf die Ischämie-Reperfusionstoleranz haben können. Ziel dieser Arbeit war es, die Wirkung einer AGE-Induktion mittels Glyoxals, ein AGE-Bildner und Dicarbonyl, im Ischämie-Reperfusionsversuch am humanen Myokard ex vivo und dessen potentielle intrazelluläre Wirkweise zu untersuchen. Dazu wurden Vorhoftrabekel junger (≤ 60 Jahre) und alter (≥ 70 Jahre) Patienten mit Glyoxal behandelt. Die Ergebnisse zeigen, dass Glyoxal konzentrationsabhängig das Myokard vor Ischämie-Reperfusionsschäden schützt. Eine protektive Wirkung infolge einer reduzierten RAGE(Rezeptor für AGE)-Expression und der Aktivierung von Survival-Kinasen wird diskutiert. Der unerwartet positive Effekt einer Proteinglykierung im Ischämie-Reperfusionsversuch könnte daher eine Grundlage für einen neuartigen Präkonditionierungsansatz sein.-
dc.description.abstractAccumulation of age-related protein modifications by carbohydrates (advanced glycation endproducts, AGEs) is discussed to have a pivotal role in the aetiology of cardiovascular diseases. Aging of the human myocardium is characterized by structural as well as functional changes that may affect the ischemia reperfusion tolerance. The aim of this study was to analyse the effect of an AGE induction by glyoxal, a dicarbonyl known to form AGEs, during experimental ischemia and reperfusion of human myocardium ex vivo and its potential intracellular influence. Therefore, right atrial trabeculae of adult (≤ 60 years) and elderly (≥ 70 years) patients were treated with glyoxal. The results reveal that Glyoxal protects the myocardium against ischemia reperfusion injury. A protective effect due to decreased RAGE (receptor for AGE)-expression and the activation of survival kinases is discussed. The unexpected positive effect of protein glycation during the reperfusion phase may be a novel approach to preconditioning.eng
