Abstract: Ziel dieser Arbeit war der Nachweis von microRNAs im Seminalplasma sowie die Analyse von möglichen Unterschieden in der Expression bei Patienten mit abnormalen Spermiogrammparametern im Vergleich zu Probanden mit Normozoospermie. In Microarrays basierend auf dem Northern Blot System wurde die Expression von 244 microRNAs des Seminalplasmas bei drei verschiedenen Teilnehmern (Normo-, Oligo-, Azoospermie) analysiert, 57 microRNAs waren zwischen den Probanden differentiell exprimiert, die Expression von 60 microRNAs war nicht detektierbar. Bei einer qPCR-Analyse der Expression der microRNAs miR-143, miR-210 und miR-375 im gesamten Kollektiv (n=94; n=50 mit Normozoospermie, n=44 mit abnormalem Spermiogramm) und Assoziation zu den Spermiogrammparametern konnte lediglich ein statistischer Trend (p=0,06) zwischen miR-375-Expression und Samenqualität gezeigt werden. Zusammenfassend liegt eine differentielle Expression von microRNAs im Seminalplasma von Kinderwunschpatienten und gesunden Probanden vor, allerdings konnte keine Assoziation zwischen der Expression der miR-143, miR-210 oder miR-375 und den klassischen Spermiogrammparametern festgestellt werden.
Aim of this study was the detection of microRNAs in seminal plasma and the analysis of their expression in patients with abnormal semen parameters compared to subjects with normozoospermia. We evaluated the expression of 244 microRNAs in the seminal plasma of a normozoospermic, an oligozoospermic and an azoospermic male by microArrays based on the Northern Blot system. 57 microRNAs were differentially expressed among the samples; 60 microRNAs were not detectable. Following, miR-143, miR-210 and miR-375 were analyzed in the study population (n= 94; n=50 with normozoospermia), n=44 with abnormal semen parameters) by qPCR. A statistical trend (p= 0,06) correlation was shown for miR-375 expression and semen quality. In conclusion, our study shows differences in the expression of microRNAs in seminal plasma of healthy males and infertility patients. Nevertheless, there was no statistical significant correlation of the expression of miR-143, miR-210 and miR-35 to classical spermiogram parameters.
