Abstract: Die Augenentwicklung im Vertebraten beginnt mit dem Auftreten von Augenbläschen auf beiden Seiten des ventralen Dienzephalons, die primär aus einem einheitlichen Pax6-positiven Augenfeld stammen. Obwohl pax6 in dem gesamten Augenfeld exprimiert wird, wird seine Expression durch die reziproke transkriptionale Repression von pax6 und pax2 auf den Augenbecher beschränkt. Es bleibt jedoch unklar, wie Pax6-Protein rechtzeitig aus dem Augenstiel-Gebiet entfernt wird. Hier berichten wir, dass Mid1 mit Pax6 interagiert. Es wird die Expression von Midline1 während der Augenentwicklung im Vergleich zu Pax2 und Pax6 dargestellt. Experimente, die die Funktion von Midline1 supprimieren zeigen, dass es zu einer Zunahme der Pax6-Expression kommt sowie zur Entwicklung eines spezifischen Augenphänotyps. Auf Proteinebene lassen sich vermehrt Pax6-positive Zellen innerhalb der Retina darstellen. Aus diesen Untersuchungsergebnissen wäre Pax6 als Substrat der Ubiquitin-Ligase Midline1 vorstellbar.
The first morphological sign of vertebrate eye formation is the appearance of eye vesicles on both sides of the ventral diencephalon, which primarily originate from a uniform Pax6-positive eye field. Although pax6 is expressed in the entire eye field, its expression is restricted to the eye cup by the reciprocal transcriptional repression of pax6 and pax2. However, it remains unclear how Pax6 protein is removed from the eyestalk territory on time.Here we report that Mid1 interacts with Pax6. The expression of Midline1 during eye development is compared to the Expression pattern of Pax2 and Pax6. Loss-of-function-experiments of Midline1 show that there is an increase in Pax6 expression as well as the development of a specific eye phenotype. On protein levels we find significantly more Pax6-positive cells within the retina. Pax6 as a substrate of the ubiquitin ligase Midline1 would be conceivable from these investigations.
