Abstract: Pankreas Stellatumzellen (PSCs) haben in den letzten Jahren immer mehr Bedeutung bei der Entstehung von Erkrankungen des Pankreas erlangt. Vor allem in Pankreaskarzinomen sind die Funktionen dieser Zellart bei der desmoplastischen Reaktion essentiell. Sie können in einen aktivierten Zustand gelangen, in welchem sie Funktionen wie die Produktion von Kollagen Typ I und III erlangen, die Apoptose der Tumorzellen hemmen und die Migration von T-Zellen reduzieren. Mittels einer Next Generation Sequenzierung wurde das Genom der Stellatumzellen von 5 Patienten untersucht, als Referenz wurden Blutproben genommen. Zur Filterung somatischer Mutationen wurde die Software MuTect verwendet. Die Daten wurden weiter mittels der Pathogenitäts-Scores SIFT, Polyphen 2, CADD und dem Mutationtaster Score analysiert. Final wurden vier Mutationen in den Genen SERPINB2, MIGA2, TOP2B und ZDHHC18 gefunden. Nach einer Visualisierung mittels IGV Viewer wurden die Mutationen durch Sanger Sequenzierungen reproduziert. Zusätzlich wurden 25 weitere Proben untersucht. Die vier ausgewählten Mutationen konnten in einer von 30 Proben reproduziert wurden. Die große Anzahl an verschiedenen Veränderungen im Genom der Stellatumzellen legt nahe, dass jeder Patient ein eigenes Mosaik an Mutationen aufweist. Die Untersuchung der funktionellen Konsequenz der Mutationen in folgenden Experimenten ist essentiell. Möglicherweise stellen sie ein potentielles neues Therapie Ziel dar.
Pancreatic stellate cells are the rising stars in pancreatic diseases, especially pancreatic cancer. In pancreatic cancer carcinogenesis, their function concerning the desmoplastic reaction is crucial. When activated PSCs get functions such as the production of collagen I and III leading to hypoxic conditions in the tumor microenvironment. Moreover, they inhibit the apoptosis of tumor cells and reduce the migration of t-cells in pancreatic tumors. The present dissertation study investigated through a Next-Generation- Sequencing (NGS) the genome of pancreatic stellate cells. The DNA of five PSC samples from pancreatic cancer patients was investigated. As a reference for a normal genome blood samples of the patients were taken. For the detection of somatic mutations, the software MuTect was used. Data were filtered using the pathogenicity prediction scores SIFT, Polyphen2, CADD, and the MutationTaster Score. Finally, four mutations in the SERPINB2, MIGA2, TOP2B, and ZDHHC18 were found. After visualization by using the Integrative Genomics Viewer (IGV), mutations were reproduced by Sanger technique. Additionally, DNA of 25 patients suffering from pancreatic cancer and pancreatitis was isolated and investigated as well. The four mutations of focus were reproduced in one of 30 samples. The high variability of single nucleotide variants in the PSCs genome leads to the conclusion that every patient has got a single mosaic of variants in their PSCs genome. The functional analysis of the mutations found in this study is crucial in further studies. PSCs might be a potential target for new treatments in pancreatic cancer patients in the future.
