dc.description.abstractAnalytische Produktauthentifizierung ist relevant, da Lebensmittelbetrug aufgrund der Unvollkommenheit der derzeit verwendeten Methoden oft nicht erkannt wird. Die epigenetische (auf DNA-Methylierung basierende) Produktauthentifizierung in biologisch und konventionell angebauten Kartoffeln (Solanum tuberosum) unter Verwendung eines Post-Bisulfit-Adapter-Tagging Whole-Genome Bisulfit-Sequencing (PBAT-WGBS) Protokolls wird in dieser Dissertation behandelt. Die Arbeit gliedert sich in drei Teile: einen methodischen Überblick über die wichtigsten Schritte, die derzeit im Bereich der DNA-Methylierungsanalyse angewandt werden, eine bioinformatische Bewertung von Alignment-Tools und die beispielhafte Anwendung der vorgeschlagenen Methode bei Kartoffeln. Neben dieser Anwendung bietet sie die Möglichkeit Lagerungs-, Transport-, Verarbeitungs- sowie weitere Anbaubedingungen in einer Vielzahl an Produkten zu analysieren. Daher kann sie in der zukünftigen Produktauthentifizierung von Bedeutung sein.ger
dc.description.abstractThe topic of product authentication is of importance as food fraud could, up to now, often not be detected due to the imperfection of currently used methods. The question of epigenetic (DNA methylation-based) product authentication in organic and conventional grown potatoes (Solanum tuberosum) using a Post-Bisulfite-Adapter-Tagging Whole-Genome Bisulfit-Sequencing (PBAT-WGBS) protocol is addressed in this dissertation. The thesis is divided into three parts: a methodological review describing core steps currently applied in the field of DNA methylation analysis, a bioinformatical evaluation of alignment tools and the exemplary application of the proposed method in potatoes grown under organic and conventional farming systems. Besides this application, it offers the potential to analytically monitor storage, transport, cultivation and processing conditions of multiple kinds of products and therefore might be of importance in future product authentication.eng
