Abstract: Thymus vulgaris, der Echte Thymian, steht exemplarisch für eine Vielzahl an Arzneipflanzen, deren ätherische Öle unter anderem in der pharmazeutischen Industrie von Bedeutung sind. Thymian prägt in seiner Wildform sechs verschiedene Chemotypen aus, die unterschiedliche Monoterpenalkohole im ätherischen Öl enthalten: Geraniol (G), α-Terpineol (A), trans-Sabinenhydrat (U), Linalool (L), Carvacrol (C) und Thymol (T). Deren Vererbung erfolgt über mindestens 5 verschiedene Allele in einer epistatischen Reihe. In der vorliegenden Arbeit wurde die Methylierung als möglicher Mechanismus der Regulation der Ausprägung der Chemotypen betrachtet, mit besonderem Fokus auf RdDM (engl. RNA-directed DNA methylation). Dazu wurde die Familie der Argonautenproteine (AGO) in Thymian als Kandidaten für diese Regulation näher untersucht. Diese Proteinfamilie wurde zuvor nicht in T. vulgaris beschrieben und spielt möglicherweise eine wichtige Rolle bei der Ausprägung der Chemotypen.
Thymus vulgaris, also known as common thyme or garden thyme, produces essential oils that are of use for the pharmaceutical industry. It is known to differentiate into six chemotypes in wild type that are each dominated by a specific monoterpene alcohol: geraniol (G), α-terpineol (A), trans-sabinene hydrate (U), linalool (L), carvacrol (C) und thymol (T). The inheritance of the chemotypes is regulated via at least five different alleles and form an epistatic row. In this work, methylation was implicated in the the control of biosynthetic genes and formation of the chemotypes, suggesting that RdDM (RNA-directed DNA methylation) may be an important mechanism. Therefore a key component of RdDM, the family of the Argonaute proteins (AGO) was investigated. This protein family has not yet been described in Thymus vulgaris and may play an important role in gene expression control in this model system.
