dc.description.abstract | Die Zytoskelett-Komponente Aktin reguliert die nukleäre Genexpression zusammen mit dem Serum Response Factor (SRF) und myocardin-related transcription factors (MRTFs). MRTF-A wird durch die Aktin-Dynamik und die Dissoziation repressiver G-Aktin:MRTF Komplexe gesteuert. WH2-Domänen von nucleation promoting factors (NPFs), wie N-WASP, WAVE2 und JMY, binden Aktin an der gleichen Stelle wie MRTF-A, was auf eine Bindungskonkurrenz deutet. In Maus-Fibroblasten konnte gezeigt werden, dass die WH2-Domänen von N-WASP und WAVE2 MRTF-A vom Aktin verdrängen und somit MRTF-A unabhängig von einer veränderten Aktin-Dynamik aktivieren. Immunfluoreszenz-Studien belegen, dass die Überexpression von N-WASP und WAVE2 zu Kern-akkumuliertem MRTF führt. Reporter-Assays zeigen eine Aktivierung von SRF durch N-WASP, WAVE2 und JMY-Mutanten. Eine gleichzeitige Arp2/3-Inhibierung oder Mutation der Arp2/3-interagierenden CA-Region kann diese NPF-vermittelte MRTF-Aktivierung nur wenig reduzieren. Diese Arbeit trägt dazu bei, die aktivierenden Mechanismen von WH2-enthaltenden Proteinen auf Aktin-MRTF-SRF-vermittelte Genexpression besser zu verstehen. | - |
dc.description.abstract | The cytoskeleton component actin regulates nuclear gene expression together with the serum response factor (SRF) and myocardin-related transcription factors (MRTFs). MRTF-A activation is restricted to alterations in actin dynamics and requires dissociation of repressive G-actin:MRTF-A complexes. WH2 domains of nucleation promoting factors (NPFs), such as N-WASP, WAVE2 and JMY, were already shown to bind actin at the same surface like MRTF-A, implicating a mutually exclusive binding. In mouse fibroblast we could show that WH2 domains of N-WASP and WAVE2 directly compete with MRTF-A for actin binding thereby activating MRTF-A independent of changes in actin dynamics. Immunofluorescence studies revealed that overexpression of N-WASP and WAVE2 lead to nuclear translocation of MRTF-A. Reporter assays demonstrated SRF activation by N-WASP, WAVE2 and JMY mutants. Simultaneous inhibition of Arp2/3 or mutation of the Arp2/3-interacting CA region only partially reduces MRTF-A activation by NPFs. This approach will help understanding the activating mechanism of WH2-containing proteins on actin-MRTF-SRF-mediated target gene transcription. | eng |
