Abstract: Das humane FK506-bindende Protein 38 (FKBP38) ist eine Peptidyl-Prolyl cis/trans Isomerase (PPIase), die eine zentrale Rolle bei der neuronalen Apoptose spielt. Das konstitutiv inaktive FKBP38 ist die erste PPIase, die von Calcium (Ca2+) und Calmodulin (CaM) reguliert wird, da nur der aktive FKBP38/CaM/Ca2+ Komplex FKBP Aktivität aufweist und den anti-apoptotischen Effektor Bcl-2 inhibieren kann. Ziele dieser Arbeit waren die Strukturaufklärung der katalytischen FKBP Domäne von FKBP38 sowie die Untersuchung der FKBP38-Wechselwirkungen mit Ca2+- CaM. Die NMR-Struktur von FKBP3835-153 weist die typische "half-barrel" Faltung der FKBP Domänen auf. Strukturelle Unterschiede zum prototypischen FKBP12 erklären die fehlende FKBP Aktivität. Außerdem wurde zum ersten Mal an einer FKBP Domäne eine Ca2+-Bindungsstelle entdeckt. Untersuchungen der FKBP38/CaM Wechselwirkungen anhand von Veränderungen der chemischen Verschiebungswerte (CSP) zeigen eine Ca2+-abhängige Bindung zwischen den C-terminalen Domänen von FKBP38 und CaM, sowie eine Ca2+-unabhängige Wechselwirkung zwischen den N-terminalen Domänen beider Proteine, welche die FKBP38 Aktivierung erklärt. Hieraus resultierten Strukturmodelle für die FKBP38/CaM/Ca2+ Komplexe bei unterschiedlichen Ca2+-Stöchiometrien, anhand derer sich eine neue Form der Enzymaktivierung durch CaM ableiten ließ.
The human FK506-binding protein 38 (FKBP38) is a peptidyl prolyl cis/trans isomerase (PPIase) that plays a key role in neuronal apoptosis. This constitutively inactive protein is the first PPIase that was found to be regulated by calmodulin (CaM) and calcium (Ca2+), as only the FKBP38/CaM/Ca2+ complex displays FKBP activity and can inhibit the anti-apoptotic effector Bcl-2. The aims of this work are the structural characterization of the catalytic FKBP domain of this protein and the study of the interactions of FKBP38 with Ca2+-CaM. The solution structure of FKBP3835-153 displays the typical "half-barrel" fold of FKBP domains. Specific differences compared to the prototypic FKBP12 explain the lack of FKBP activity of FKBP38. Moreover, for the first time a Ca2+-binding site was found in an FKBP domain. Chemical shift perturbation (CSP) studies of the FKBP38/CaM interactions demonstrate Ca2+-dependent binding between the Cterminal domains of FKBP38 and CaM, as well as a Ca2+-independent interaction between the respective N-terminal domains that explains the activation of FKBP38. Structural models were derived for the FKBP38/CaM/Ca2+ complexes at different Ca2+ stoichiometries, thereby revealing a novel scenario of target activation by CaM.
