Abstract: Die hochauflösende Ultraschallmikroskopie gewinnt in der medizinischen Forschung zunehmend an Bedeutung. In der vorliegenden Arbeit wurden mit Ascorbinsäure (50 µg/ml) und Vitamin D3 (10 nmol/l) sowie der Kombination beider Substanzen stimulierte humane Osteoblasten mit dem akustischen Rastermikroskop KSI SAM 2000 bei einer Arbeitsfrequenz von 900 MHz untersucht. Mit Hilfe des Cell-Analysis-Verfahrens wurde ein quantitatives Auswertungsverfahren zur Bestimmung der morphologischen und akustischen Parameter Zelldicke (d), Schallgeschwindigkeit und des Schallschwächungskoeffizienten in der nukleären, perinukleären und peripheren Zone erstmals an humanen Osteoblasten angewandt. Es wurde festgestellt, dass das Cell-Analysis-Verfahren eine sichere Zelldickenbestimmung für d > 1 µm gestattet. Die deutlichsten Veränderungen der Zellparameter traten in der perinukleären Zone auf. Dabei verminderte sich bei Stimulation mit Vitamin D3 die Zelldicke um 37 % und die Schallgeschwindigkeit erhöhte sich um 8 %. Der Schallschwächungskoeffizient nahm bei der Kombinationsstimulation um 88 % zu. Mit konfokaler Laserscanningmikroskopie wurde vergleichend gezeigt, dass der Kalziumgehalt in der perinukleären Zone der stimulierten Osteoblasten am größten ist. Weiterhin wurden mit allen verwendeten mikroskopischen Verfahren (Lichtmikroskopie, Fluoreszenzmikroskopie, akustische Rastermikroskopie) in der perinukleären Zone Granulationen sowie eine Veränderung der Form und Größe der Granula bei den Stimulationen beobachtet. Eine signifikante Abhängigkeit der Zellparameter von der Kulturdauer konnte nicht festgestellt werden. Es kann vermutet werden, dass die Veränderungen der Zellparameter sowohl durch strukturelle Änderungen als auch durch eine Änderung des Kalziumgehalts vor allem im Endoplasmatischen Retikulum verursacht wurden. Mit der akustischen Rastermikroskopie steht daher in Kombination mit dem Cell-Analysis-Verfahren ein leistungsfähiges und weitgehend rückwirkungsfreies Instrument zur quantitativen Bestimmung morphologischer und akustischer Zellparameter in vitro zur Verfügung.
The high-resolved ultrasonic microscopy obtains increasing significance in the medical research. In the present study stimulated human osteoblasts (ascorbic acid 50 µg/ml, vitamin D3 10 nmol/l and the combination of both substances) were examined with the Scanning Acoustic Microscope KSI SAM 2000 at 900 MHz. With the the Cell-analysis-method a quantitative tool was applied first time for determining morphological and acoustical cell parameters. Thickness thickness (d), the speed of sound and the attenuation coefficient were determined in the nuclear, perinuclear and peripheral zone of human Oosteoblastens. It was found out that the Cell-analysis-method allows the cell thickness measurement for d > 1 µm. Significant changes of the cell parameters were observed in the perinuclear zone. The cell thickness decreased under stimulation with vitamin D3 for about 37 % and the speed of sound increased by 8 %. The attenuation coefficient increased under the combined stimulation for about 88 %. By Confocal Laser Scanning Microscopy it was shown that the calcium content is largest in the perinuclear zone of the stimulated osteoblasts. Furthermore, the appearance of granulates was observed in the perinuclear zone with different microscopic methods (light microscopy, fluorescence microscopy, acoustic microscopy). A significant dependence of the cell parameters on the cultural duration could not be proved. It can be assumed that the changes of the cell parameters could be caused by structural variations and by the modification of the calcium content especially of the endoplasmic reticulum. The Scanning Acoustic Microscopy in combination with the Cell-analysis-method is a powerful and non-violating tool for the quantitative measurement of morphological and acoustical parameters of cells in vitro.
