Abstract: Hintergrund: Die Fähigkeit zur Fieberausbildung während einer schweren bakteriellen Infektion scheint essentiell für das Überleben des Erkrankten zu sein. Beim Menschen ist es nach wie vor unklar, ob Fieber im Rahmen der frühen nichtadaptiven Immunantwort einen vorteilhaften Effekt auf die Expression proinflammatorischer Zytokine ausübt. Methode: Ziel dieser Studie war es, den Einfluss von Normo- und Hyperthermie auf die intrazelluläre Expression proinflammatorischer Zytokine (IL6, IL8, TNFα) mit Hilfe der Durchflusszytometrie zu untersuchen. Hierzu wurden Vollblutproben junger gesunder Männer verwandt. Lipopolysaccharid- stimulierte (10 ng/ml) und nicht- stimulierte Monozyten wurden zu verschiedenen Zeitpunkten (1, 2, 3, 6 h) unterschiedlichen Temperaturen ausgesetzt (37, 39, 42°C). Resultate: Temperatur bewirkt eine Zytokinexpression in Monozyten nur in Anwesenheit von Lipopolysaccharid (LPS). Hyperthermie und LPS modulieren die Zytokinproduktion in unterschiedlicher Art und Weise. Die Expression von IL8 und TNFα stagniert nach 3 h Stimulation (p Zusammenfassung: Es ist bekannt, dass eine prolongierte oder exazerbierte Expression von IL8 und TNFα zu Zytotoxizität und Letalität des Organismus führt. Eine Weiterführung der IL6-Expression während der frühen nichtadaptiven Immunantwort ist wichtig für die Induktion der nachfolgend einsetzenden Akuten Phase Reaktion. Der Einfluss von Hyperthermie scheint sowohl zytoprotektiv auf bakteriell stimulierte Zellen der frühen nichtadaptiven Immunantwort zu wirken, als auch von Vorteil für die Weiterführung der späten Immunantwort zu sein.
Background: The ability to develop fever during a severe bacterial infection seems to be essential for survival. In humans it is unclear, weather fever in the course of nonadaptive immune response has a favourable effect on the expression of pro-inflammatory cytokines. Methods: The aim of the study was to investigate the influence of normo- and hyperthermia on the intracellular expression of proinflammatory cytokines (IL6, IL8, TNFα) by flow cytometry. Whole blood assays of young healthy men were used. Monocytes stimulated or not stimulated with lipopolysacccharide (10 ng/ml) were treated at different time points (1, 2, 3, 6 hours) with temperature (37, 39 and 42°C). Results: Temperature provokes cytokine expression in monocytes only in the presence of LPS. Hyperthermia and LPS modulate cytokines differently. The expression of IL8 and TNFα stops after 3 hours of stimulation (p Conclusions: It is well known that an unrestrained expression of IL8 and TNFα leeds to cytotoxicity and lethality. A continuing production of IL6 during the nonadaptive immune response is important for the induction of the acute phase. Hyperthermia seems to be advantageable for the balance of cytokine expression and beneficial for the initiation of late immune answer.
