Abstract: Xanthomonas campestris pv. vesicatoria (Xcv) transloziert mindestens 20 Effektorproteine durch ein spezialisiertes Typ-III-Sekretionssystem in die pflanzliche Wirtszelle. AvrBs1, AvrBs3 und AvrBs4 sind drei dieser Typ-III-Effektoren, welche aufgrund ihrer Fähigkeit, in resistenten Pflanzen eine HR auszulösen, als Avirulenzproteine bezeichnet wurden. In der vorliegenden Arbeit wurden deren Primärfunktionen als Virulenzfaktoren des Pathogens untersucht. Im Vordergrund stand die Analyse von AvrBs3, einem der bestuntersuchten Typ-III-Effektoren aus Xcv. Im Mittelpunkt stand dabei die Identifizierung und Analyse von Proteinen, die mit AvrBs3 interagieren. Es wurde gezeigt, dass AvrBs3 mit dem Xcv-Typ-III-Chaperon HpaB interagiert, welches die Sekretion des Effektorproteins begünstigt. Weitere Untersuchungen zeigten, dass AvrBs3 im Zytoplasma der Pflanzenzelle in Abhängigkeit von der zentralen "Repeat"-Region Homodimere bildet. Die Interaktion zwischen AvrBs3 und Importin α1 aus Capsicum annuum (CaIMPα1), welches den Kernimport des Effektors vermittelt, wurde in planta gezeigt. In Hefe-Di-Hybrid-Sichtungen einer cDNA-Bank aus Lycopersicon esculentum mit AvrBs3 als Köderprotein wurden zwei interessante, pflanzliche Proteine identifiziert, welche potentielle Virulenzzielproteine von AvrBs3 darstellen und eine Rolle für die Virulenz von Xcv spielen könnten: Der Interaktor LeThiC ist vermutlich an der Thiaminbiosynthese beteiligt und Aip8 ("protein interacting with AvrBs3 8) zeigt Ähnlichkeit zu einem Transkriptionsregulator aus Arabidopsis. Mikroskopische Studien nach Agrobacterium vermittelter Expression von avrBs3 in Nicotiana benthamiana zeigten, dass AvrBs3 eine erhöhte Transkriptionsaktivität in den Zellkernen bewirkt. Vergleichende mikroskopische Analysen von Pflanzenzellen, die avrBs3, avrBs1 oder avrBs4 exprimieren, ergaben außerdem, dass AvrBs1 eine ähnliche Vergrößerung von Mesophyllzellen induziert wie AvrBs3. Physiologische Untersuchungen zeigten, dass AvrBs1 und AvrBs3 eine Erhöhung des Ionenaustritts aus N. benthamiana-Zellen bewirken. Das AvrBs3-Familienmitglied AvrBs4 induziert dagegen weder eine starke Zellvergrößerung noch einen verstärkten Ionenaustritt, jedoch eine Erhöhung des Katalasegehaltes in Peroxisomen. Des Weiteren wurde beobachtet, dass AvrBs3 eine supprimierende Wirkung auf die durch AvrBs1 induzierte HR in Bs1-enthaltenden Paprikapflanzen hat.
Xanthomonas campestris pv. vesicatoria (Xcv) translocates at least 20 effector proteins through a specialized type III secretion system into the plant host cell. Among these are AvrBs1, AvrBs3 and AvrBs4, type III effectors that were termed avirulence proteines because of their ability to induce the HR in resistent plants. In the present work, the primary functions of these effectors were studied which are proposed to enhance the virulence of the pathogen. The main focus of this work was the analysis of AvrBs3, a well-studied type III effector of Xcv. The central question dealt with the identification and analyses of proteins interacting with AvrBs3 (Aip). First, the interaction of AvrBs3 with HpaB, a type III chaperone which promotes the secretion of the effector, was shown. Secondly, the present analyses revealed that AvrBs3 homodimerizes in the plant cell cytoplasm, mediated by the central repeat region of the effector. The interaction of AvrBs3 with Importin α1 from Capsicum annuum (CaIMPα1), which mediates the import of the effector into the plant cell nucleus, was demonstrated in planta. In yeast two-hybrid screens using AvrBs3 as bait two interesting proteins from Lycopersicon esculentum were identified displaying potential virulence targets of AvrBs3: LeThiC, probably an enzyme of the thiamine synthesis, and Aip8, a putative transcriptional regulator. Furthermore, microscopic studies following Agrobacterium-mediated transient expression of avrBs3 in Nicotiana benthamiana revealed that AvrBs3 causes enhanced transcription activity in plant cell nuclei. Comparative microscopic analyses of cells expressing avrBs3, avrBs1 or avrBs4 showed that AvrBs1 induces a similar enlargement of mesophyll cells as AvrBs3. Physiological analyses revealed that AvrBs1 and AvrBs3 also cause increased ion leakage in N. benthamiana. However, the AvrBs3 family member AvrBs4 showed almost no effect on cell size or ion leakage, but induces an increased content of catalase in peroxisomes. Furthermore, a cell death suppressing activity of AvrBs3 was observed in Bs1 pepper plants after delivery of AvrBs3 and AvrBs1 by Xcv.
