Zusammenfassung: In der Biosynthese der Opiumalkaloide in Schlafmohn spielen O-Methyltransferasen eine wichtige Rolle. Keimlingshypokotyle wurde mit einem sense-Konstrukt der (S)-Retikulin-7-O-Methyl-transferase (7OMT) transformiert. Aus Kalluskulturen konnten durch somatische Embryogenese transgene Pflanzen generiert und bis in die T2-Generation untersucht werden. Northern-Blot-Analysen zeigten die Überexpression der 7omt. Die Latexalkaloide wurden mittels HPLC qualitativ und quantitativ bestimmt. Die 7omt-Überexpression hatte kaum Einfluss auf die Mengen des Substrates Retikulin und der Produkte Laudanin und Laudanosin, allerdings war die Synthese der Endmetabolite des Morphinanweges beeinflußt. Die Pflanzen zeigten starke Änderungen bis zu 28 % am Gesamtalkaloidgehaltes für Morphin, 22 % für Codein, 32 % für Thebain und 9 % für Oripavin. Es gab kein einheitliches Muster, aber eine häufig auftretende antagonistische Verschiebung der Werte für Morphin/Codein gegen die Werte für Thebain/Oripavin war. An der Synthese sind außerdem zwei N-Methyltranferasen, die (S)-Coclaurin-N-Methyltransferase (CNMT) und die (S)-Tetrahydroprotoberberin-cis-N-Methyltransferase (SNMT), beteiligt, deren Sequenzen bisher aber nicht bekannt waren. Durch RT-PCR konnte eine cDNS mit Homologie zu der bereits aus C. japonica bekannten CNMT isoliert und heterolog exprimert werden. Das Enzym akzeptierte (R)- und (S)-Coclaurin, (S)-Norretikulin, (S)-Nororientalin und (R,S)-Norlaudanosolin als Substrate. Ausgehend von einer 135 bp umfassenden cDNS, ebenfalls mit Homologie zur CNMT aus C. japonica, gelang es, mittels RACE-PCR zwei Klone einer bisher unbekannten cDNS zu isolieren. Rohextraktmessungen zeigten Aktivität beider Klone bei der N-Methylierung von (S)-Stylopin. Diese Ergebnisse wurden massenspektrometrisch bestätigt. Es handelte sich um die bis dato noch nicht bekannte Sequenz der (S)-Tetrahydroprotoberberin-cis-N-Methyltransferase, die (S)-Stylopin im Benzophen[c]anthridinzweig der Alkaloidbiosynthese in P. somniferum umsetzt.
O-Methyltransferases play an important role in the biosynthesis of alkaloids in opium poppy. Hypokotyls of seedlings were transformed with a sense construct of (S)-reticuline 7-O-methyltransferase (7OMT). Transgenic plants could be generated by somatic embryogenesis from callus cultures and were investigated through T2 generation. Northern-Blot-Analyses showed the 7omt overexpression. Latex alkaloids were qualitatively and quantitatively determined by HPLC. The 7omt overexpression had only minor influence on the amounts of the substrate reticuline and the products laudanine and laudanosine, but on the end products of the morphinane branch. The plants showed changes up to 28 % of the total alkaloid content for morphine, 22 % for codeine, 32 % for thebaine and 9 % for oripavine. There was no consistent pattern but a frequent antagonistic shift of the values of morphine/codeine against thebaine/oripavine. Additionally there are two N-methyltransferases, the (S)-coclaurine N-methyltransferase (CNMT) and the (S)-tetrahydroprotoberberine cis-N-methyltransferase (SNMT) in the biosynthesis, which sequences were not known at that point. By RT-PCR a cDNA homolog to the CNMT of C. japonica could be isolated and heterologous expressed. The enzyme accepted (R)- and (S)-coclaurine, (S)-norreticuline, (S)-nororientaline and (R,S)-norlaudanosoline as substrates. Coming from a 135 bp cDNA with homology to the CNMT of C. japonica two clones of an unknown cDNA could be isolated by RACE-PCR. Measurements with bacterial lysates showed activity for both clones in N-methylating (S)-stylopine. The results could be approved by mass spectrometry. It was shown that the isolated sequences coded for the so far unknown sequence of the (S)-tetrahydro-protoberberine cis-N-methyltransferase that N-methylates (S)-stylopine in the benzophenanthridine branch of the alkaloid biosynthesis in P. somniferum.
