dc.description.abstractDer Schlafmohn Papaver somniferum, eine der ältesten Kulturpflanzen der Welt, akkumuliert Benzylisochinolinalkaloide. Diese sich von der Aminosäure L-Tyrosin ableitenden Alkaloide kommen neben den Mohngewächsen schwerpunktmäßig in der Pflanzenordnung der hahnenfußartigen Gewächse vor. Dementsprechend lieferten der Schlafmohn und eine Berberitze (Berberis stolonifera) das Material für die vorliegende Arbeit. Die Biosynthese der Benzylisochinolinalkaloide ist auf enzymatischer Ebene weitgehend aufgeklärt. Bis zum (S) - Retikulin sind des Weiteren die DNA Sequenzen der Enzyme bekannt. Für die Biosynthesen der einzelnen Alkaloidklassen ist die Biosynthese der Berberine fast vollständig auf DNA-Ebene geklärt. Die Biosynthese der Benzophenanthridine, wie die der Morphinane, ist Gegenstand derzeitiger Untersuchungen. Die vorliegende Arbeit vervollständigt mit der Isolierung der cDNA einer (S) - Tetrahydroprotoberberinoxidase die Informationen für die Synthese der Berberine. Für die Biosynthese der Morphinane wird die Salutaridinsynthaseaktivität des CYP719B1 nachgewiesen. Damit ist ein Enzym mit hohem biotechnologischen Potential erstmals rekombinant erhältlich und somit gezielten Modifikationen und Mutationen zugänglich.-
dc.description.abstractThe Opium poppy papaver somniferum, one of the world oldest cultivated plants, is accumulating benzylisoquinolines. Besides the Papaverales this L-Tyrosin derived alkaloids are occurring frequently in plants of the order Ranunculales. Accordingly the raw material for the investigations are provided by the opium poppy and a barberry variety (Berberis stolonifolia). The enzymatic reactions of benzylisoquinolins biosynthesis are well investigated. The biosynthesis leading to (S)-reticuline is elucidated on the DNA level. Sequences are also known for nearly all biosynthetic steps of Berberine biosynthesis. The Morphinane and Benzophenanthridine biosynthesis are under current investigation. Results of the study presented here includes the identification of the DNA-Sequence of a (S)-tetrahydroprotoberberine oxidase, the missing step in the Berberine biosynthesis. In Morphinane biosynthesis the salutaridine synthase activity of CYP719B1 is shown. Therefore an enzyme with high potential in biotechnology application is recombinant expressed for the first time and is hence available for targeted mutations and modifications.eng
