dc.description.abstractDie reversible Proteinphosphorylierung an Serin- oder Threoninresten N-terminal zum Prolin gehört zu den wichtigsten Regulationsmechanismen einer Vielzahl zellulärer Prozesse. Die Phosphorylierung wird durch die Vertreter der prolinspezifischen Proteinkinasen katalysiert. Diese werden in die Familien der cyclinabhängigen Kinasen, welche eine Schlüsselrolle bei der Regulation des Zellzyklus besitzen, und in die der Mitogen-aktivierten Proteinkinasen, die entscheidend an der intrazellulären Signalübertragung beteiligt sind, eingeteilt. Durch die einzigartigen Eigenschaften des Prolins kann es zum gleichzeitigen Auftreten von cis- und trans-Isomeren der Peptidyl-Prolylbindung in nativen Proteinen kommen. Wie am Beispiel von Proteasen bereits gezeigt wurde, können diese sich ineinander umwandelnden Konformere in Enzymreaktionen unterschiedlich reagieren. Im Verlauf dieser Arbeit wurde unter Verwendung von Oligopeptidsubstraten erstmals gezeigt, dass auch die Phosphorylierung von Ser/Thr-Pro Motiven durch prolinspezifische Kinasen isomerspezifisch verläuft. Sowohl die cyclinabhängige Kinase Cdc2/Cyclin B als auch die MAPK ERK2 phosphorylierten ausschließlich das trans-Konformer der Ser/Thr-Pro-Motive. Die nachgewiesene Isomerspezifität schlägt ein neuartiges Regulationsprinzip zur Kontrolle der prolinpezifischen Phosphorylierung vor. So konnte anhand des Modellproteins Ribonuklease T1 gezeigt werden, dass Peptidyl-Prolyl-cis/trans-Isomerasen (PPIasen), durch die Katalyse der Prolylisomerisierung, die Bereitstellung des phosphorylierbaren Konformers regeln können. Weiterhin zeigten die Untersuchungen zur Phosphorylierung der RNase T1, dass die isomerspezifische Phosphorylierung auch als eine neue, sehr empfindliche Methode zur Strukturaufklärung von Proteinen dienen kann.-
dc.description.abstractPhosphorylation of serine or threonine residues preceding proline is catalysed by proline-directed protein kinases. They include the multigene families of cyclin-dependent protein kinases (Cdks), which play an important role in the control of cell cycle, and of mitogen activated protein (MAP) kinases that are involved in signal transduction pathways. Unlike other peptide bonds peptidyl-prolyl bonds form cis and trans isomers of comparable thermodynamic stability. Thus, slowly interconverting prolyl isomers of native proteins offer alternatives for proteins to bind. Consequently, isomer specific reactions have already been identified in the case of proteases. By using short oligopeptide substrates we were able to demonstrate that the proline-directed MAP kinase ERK2 and the cyclin-dependent protein kinase Cdc2/Cyclin B phosphorylate exclusively the trans isomer of Ser/Thr-Pro motives. These results show that correct signature sequences are not sufficent to render potential substrates reactive to proline-directed enzymatic phosphorylations, but that the conformational state of the peptide bond following serine (threonine) is a critical determinant. Furthermore, the phosphorylation of the model protein RNase T1 proposes a novel regulatory mechanism for the control of proline-directed phosphorylation. Thus we could demonstrate that peptidyl prolyl cis/trans isomerases can facilitate the phosphorylation by catalysing cis/trans isomerization. Moreover, the discovery of isomer specific protein kinases provides a novel tool to detect small populations of co-existing trans conformers of Ser/Thr-Pro bonds that has been described as cis in the native state.eng
