Abstract: Durch ektopisch exprimierte Kresoxim-methyl Einkettenantikörper (anti-Kres-scFv) sollte die Bioverfügbarkeit des Fungizides am Wirkort nach Applikation reduziert werden. Dazu wurden Hybridomzellklone isoliert, welche spezifisch gegen Kresoxim-methyl gerichtete Antikörper sezernieren. Ausgehend von diesen Hybridomklonen wurden Einkettenantikörper regeneriert, welche spezifisch Kresoxim-methyl binden und damit theoretisch die Bioverfügbarkeit des freien Moleküls herabsetzen können. Diese scFv wurden in Tabakpflanzen zur Expression gebracht. Die regenerierten Pflanzen waren phänotypisch bei Abwesenheit von Kresoxim-methyl nicht von Wildtyp-Tabakpflanzen unterscheidbar. Unterschiede ergaben sich jedoch bei Applikation von Kresoxim-methyl während der Samenkeimung. Kresoxim-methyl greift als Strobilurinfungizid in die Atmungskette ein, hemmt den Elektronentransport am bc1-Komplex und inhibiert damit einen für die Keimung wichtigen Energiegewinnungsprozeß. Eine Fungizidabhängige Hemmung der Keimung wurde bei allen untersuchten Linien durch eine Reduktion des Frischgewichtes nach Inkubation in Kresoxim-methyl festgestellt. Die transgenen Linien mit Expression von funktionellen anti-Kres-scFv zeigten jedoch eine signifikant geringere Hemmung der Keimung. Daraus wurde geschlußfolgert, daß die Kresoxim-methyl bedingte Hemmung des Keimprozesses von Tabakpflanzen durch einen im Gewebe exprimierten, funktionellen anti-Kres-scFv vermindert wird. Desweiteren konnte das Expressionsniveau eines scFv durch Fusion mit einem Elastinartigem Polypeptid (ELP) drastisch erhöht werden. Es lag in allen Fällen deutlich (14-fach bis 230-fach) über dem Expressionsniveau nach entsprechender Expression des Einkettenantikörpers ohne ELP-Domäne. Die Affinität der scFv-Domäne für freies Kresoxim-methyl wurde durch die Fusion mit dem ELP nicht beeinträchtigt. Der Anteil des aktiven scFv am Gesamt-scFv-Gehalt war jedoch nach Expression im Samen deutlich höher als im Blatt. Die Ergebnisse zeigen, daß die Akkumulation von scFv-Protein durch die Fusion mit einem elastinartigem Polypeptid positiv beeinflußt werden kann und bei Expression in geeigneten Organen ein hoher Anteil des Proteins in aktiver Form vorliegt.
The ectopic expression of specific Kresoxim-methyl single chain antibody fragments (anti-Kres scFv) in transgenic tobacco plants was used to reduce the bio-availability of the fungicide after application. For this purpose hybridoma cell lines secreting specific antibodies against Kresoxim methyl were isolated. On the basis of this hybridoma cell lines, single chain antibody fragments were regenerated. It has been shown that the regenerated scFv`s specific binds Kresoxim-methyl and therefore theoretically the bio-availability of the free molecule can be lowered. These scFv has been expressed in tobacco plants. In the absence of Kresoxim-methyl the regenerated plants were phenotypically not distinguishable to wildtyp control-plants. Differences resulted however in the case of application of Kresoxim-methyl during the seed germination. Kresoxim-metyhl as strobilurinfungicide intervenes in the respiratory chain, restrains electron transport at the bc1-Komplex and thereby inhibit an energy production process important for germination. An fungicide-dependent inhibition of the germination process was observed in all examined lines. The transgenic lines expressing functional anti-Kres scFv however showed a significant reduced inhibition of germination. It seems that Kresoxim-methyl derived inhibition of the germination process of tobacco plants could be decreased by the expression of functional anti-Kres scFv. Furthermore the expression level of an scFv was drastically increased by the fusion with an elastinlike polypeptide. The level of expression were increased of about 14-fold to 230-fold compared to the expression level after appropriate expression of a scFv protein without ELP domain. The affinity of the scFv domain for free Kresoxim-methyl was not impaired by the fusion with the Elastin, but however the portion of the active scFv on the total scFv-content was organ-specifically and clearly higher with expression in the seed than in the leaf. The results show that the accumulation of a scFv can be positively affected by the fusion with a ELP and that after expression in suitable organs a high portion of the protein is present in active form.
