dc.description.abstractDas humane Parvulin 14 (hPar14) gehört zur Parvulin-Familie der Peptidyl-Prolyl cis/trans Isomerasen (PPIasen). Es wird angenommen, daß das Protein eine Rolle bei der Regulierung des Zellzyklus und der Remodellierung von Chromatin spielt. hPar14 ist im Zellkern lokalisiert und bindet an doppelsträngige DNA. Allerdings ist die molekulare Basis der zellulären Lokalisierung und Funktion noch unklar. In dieser Arbeit kann gezeigt werden, daß die Lokalisierung und DNA Bindung von hPar14 durch eine posttranslationale Modifikation in der N-terminalen Domäne des Proteins reguliert wird. Durch Phosphorylierung wird die Lokalisierung im Zellkern reguliert. Verschieden Kinasen der AGC-Klasse und CK2 sind in der Lage hPar14 in seiner N-terminalen Extension zu phosphorylieren. Es konnte gezeigt werden, daß eine Phosphorylierung im CK2-Erkennungsmotif eine Vorraussetzung für eine Kernlokalisierung des Proteins ist. Der Export von hPar14 ist abhängig von einer Bindung an 14-3-3 Proteine. Diese Protein/Protein Interaktion ist durch die Phosphorylierung zweier Serinreste innerhalb des 14-3-3 Bindungsmotifs in der N-terminalen Erweiterung von hPar14 reguliert.-
dc.description.abstractHuman Parvulin 14 (hPar14) belongs to the parvulin family of peptidyl-prolyl cis/trans isomerases (PPIases), and is thought to play a role in cell-cycle regulation and chromatin remodeling. Although hPar14 is nuclearly localized, and binds to double-stranded DNA, the molecular basis of the subcellular localization and the functional regulation remaines unknown. In this work it is shown that subcellular localisation and DNA-binding ability of hPar14 is regulated by posttranslational modification of its N-terminal domain. hPar14 is imported to the nucleus in a phosphorylation dependent manner. The protein is phosphorylated in the N-terminal extension by several AGC-class kinases and CK2. The phosphorylation at the CK2 recognition motif was shown to be essential for nuclear localisation of hPar14. The protein is exported from the nucleus upon binding to 14-3-3 protein. This protein/protein interaction is regulated by phosphorylation at two serine residues located within the 14-3-3 binding motif in the N-terminal extension of hPar14.eng
