dc.description.abstractDer Inhalt dieser Arbeit beschäftigt sich mit der Optimierung diagnostischer Enzyme mit Methoden des evolutionären Proteindesign. Im ersten Teil wird eine neue Methode der homologen in vitro Rekombination vorgestellt, welche aus fünf Schritten besteht: (1) Synthese Uracil-DNA-haltiger DNA (U-DNA) von den Elterngen-Sequenzen, (2) Synthese von DNA-Fragmenten mittels einer Kettenabbruchreaktion mit Didesoxynukleotiden und mit U-DNA als Template, (3) Abbau der Template-U-DNA, (4) Reinigung der DNA-Fragmente und (5) Reassemblierung der Fragmente mit gleichzeitigem Abbau der terminierenden Didesoxynukleotide. Die Funktionsfähigkeit der entwickelten Methode wurde durch Rekombination von zwei Genen der Alkalischen Phosphatase und anschließende Sequenzdatenanalyse überprüft. Der zweite Teil dieser Arbeit beschreibt die Erhöhung der thermischen Stabilität der Creatinase aus Erwinia sp. unter Beibehaltung der katalytischen Effizienz. Diese Optimierung umfasste zwei Zyklen fehlerhafter PCR mit anschließendem Screening auf thermische Stabilität und auf Enzymaktivität bei Substratlimitation. Anschließend wurden die Aminosäuren an den identifizierten Positionen permutiert. Die finale Mutante CTsd7 unterschied sich in ihrem apparenten Schmelzpunkt um 8 °C vom Wildtyp.-
dc.description.abstractThis thesis is about the optimisation of diagnostic enzymes using methods of evolutionary protein design. The first part describes the development of a new method for homologous in vitro recombination, which consists of the following five steps: (1) synthesis of uracil containing DNA from the parental sequences, (2) synthesis of DNA fragments using the principle of the Sanger sequencing method with didesoxynucleotides and with uracil containing DNA as template, (3) digestion of uracil containing template DNA, (4) purification of DNA fragments and (5) reassembly of fragments and simultaneous digestion of terminating didesoxynucleotides. The efficiency of this method has been proved by recombination of two alkaline phosphatase genes and subsequent sequence analysis. The second part describes the optimisation of creatinase from Erwinia sp. in order to achieve an increased thermal stability while catalytic efficiency had to be kept constant. This optimisation has been done by two cycles of mutagenic PCR and subsequent screening of enzyme libraries for thermal stability and for enzyme activity at limitating substrate concentrations. Positions of beneficial identified amino acids have been afterwards permutated. The apparent melting point of the final mutant CTsd7 is 8 °C increased compared to the wild type.eng
