Abstract: Die Arbeit liefert Ergebnisse zur mathematischen Modellierung von in vitro Membrandiffusionsexperimenten von hydrophilen und lipophilen Pharmaka. Dabei werden die Rolle der ungerührten Wasserschicht, die Proteinbindung und die Bildung von Mizellen im Medium untersucht. Als mathematische Modelle werden Kompartmentmodelle, eine empirische Transitzeit-Dichteverteilung (Inverse Gauss Verteilung) und eine Lösung der Membrandiffusionsgleichung (im Laplace Raum) eingesetzt. Eine Optimierung des Sampling Schemas mit Hilfe der Sensitivitätsanalyse ermöglicht eine zuverlässige Parameterschätzung auch mit weniger Messpunkten. Mit Hilfe einer Monte Carlo Simulation werden die Ergebnisse validiert. Weitere Simulationen werden mit einem Ganzkörpersimulationsprogramm durchgeführt, welches im Rahmen dieser Arbeit um eine grafische Benutzeroberfläche und die Möglichkeit verschiedener Organsimulationsfunktionen erweitert wurde.
This thesis presents results of the mathematical modelling of in vitro membrane diffusion experiments of hydrophilic and lipophilic drugs. The role of the unstirred water layer, the protein binding and the forming of micells in the medium were analysed. As mathematical models, compartmental models, an empiric transit time density distribution (inverse Gauss distribution) and a solution of the membrane diffusion equation (in the Laplace domain) were used. The optimisation of the sampling scheme with the help of the sensitivity analysis allows a reliable parameter estimation even with less data samples. The results were validated by a Monte Carlo simulation. Further simulations were performed using a whole body recirculation model which was extended by a graphical user interface and the possibility to add various organ simulation functions.
