dc.description.abstractIn Chloroplasten wird ein Teil von Thylakoidproteinen, deren Signalpeptide ein konserviertes Zwillings-Arginin-Motiv enthalten, über den ΔpH- bzw. TAT- (twin-arginine translocation) abhängigen Weg transportiert. Durch den Einsatz des chimären 16/23-Proteins, welches zwei unterschiedliche nacheinander auftretende Translokationsintermediate während des Thylakoidtransports aufweist, konnte der ΔpH/TATabhängige Transportprozess in vier Teilschritte aufgegliedert werden. Dieser Prozess wird durch eine nicht-unterstützte "loop"-Insertion des Vorläuferproteins initiiert, wobei diese unabhängig von jeglichen Proteinkomponenten der Thylakoidmembran ist. Der ΔpH/TATabhängige Transport wird durch drei Proteine der Thylakoidmembran, TatA, TatB und TatC, ermöglicht, wobei zumindest im Fall der zwei letzten festgestellt wurde, dass sie Komplexe mit Vorläuferproteinen bilden. Außerdem wurden zahlreiche Bindestellen für Vorläuferproteine in den Proteinen TatB und TatC gefunden, was darauf hinweist, dass beide Proteine Komponenten der ΔpH/TAT-abhängigen Transportmaschinerie sind.-
dc.description.abstractIn chloroplasts of higher plants, a subset of thylakoid proteins with signal peptides harbouring twin-arginine consensus motif are transported by the ΔpH- or TAT (twin-arginine translocation)-dependent pathway. By using a chimeric protein 16/23, which appears to have two distinct sequential translocation intermediates during thylakoid transport, the ΔpH/TATdependent transport process could be dissected into four steps. This process is initiated by an unassisted loop insertion of the precursor protein, which is independent of any proteinaceous components at the thylakoid membrane. ΔpH/TAT-dependent transport is facilitated by three thylakoid membrane proteins, TatA, TatB and TatC, and at least the later two proteins were found to form complexes with precursor proteins. Numerous binding sites for precursor proteins were found in TatB and TatC, suggesting that both proteins are components of the ΔpH/TAT-dependent translocation machinery.eng
