Abstract: Das Gen Lbx1 kodiert für ein Homeodomänprotein und fungiert als Transkriptionsfaktor während der Embryonalentwicklung. Zielstellung dieser Arbeit war die Untersuchung des Einflusses von Lbx1 auf die Migration diverser Vorläuferzellen und auf den Determinierungsprozess myogener und neuronaler Zellen. Im Einzelnen wurden Wechselwirkungen von Lbx1 mit anderen Genen und der Funktionsausfall dieses Genes anhand einer Lbx1 Knock-out Maus untersucht. Lbx1 spezifiziert zu einem bereits sehr frühen Zeitpunkt Muskelvorläuferzellen, die aus dem Dermamyotom der Somiten in das Mesenchym der Extremitätenknospen einwandern. Die Lbx1 Knock-out Maus zeig eine Spaltung dieser Population in zwei Subpopulationen: Zellen, die aus dem Somiten zu migrieren vermögen und Zellen, die die Fähigkeit zu einer zielgerichteten Migration verloren haben. Lbx1 spezifiziert eine Subpopulation der kardialen Neuralleistenzellen, die in das noch tubuläre Herz einwandert. Durch einen Funktionsausfall von Lbx1 ist die Migration dieser Zellen nicht gestört. Mutantenherzen zeigen aber eine Hyperplasie des Myokardiums aufgrund einer erhöhten Proliferationsrate. Bei der Entwicklung von Interneuronen des Neuralrohrs vermittelt Lbx1 positions-bestimmende determinierende Signale in einer ventral gelegenen Subpopulation dorsaler Interneurone. In Knock-out Mäusen zeigt diese Region eine Verringerung der Anzahl der Neuronen. Lbx1 Knock-out Mäuse und Pax3 Mutanten zeigen, dass beide Gene in eine negativ regulatorische Feed-back-Schleife eingebunden sind.
The gene Lbx1 encodes a homeodomain protein which functions as a transcription factor during embryonal development. The aim of this work was to investigate the influence of Lbx1 on the migration of progenitor cells and on myogenic and neuronal determination processes. In detail the interaction between Lbx1 and other genes have been investigated as well as the loss of function in Lbx1 knock-out mice. Lbx1 specifies muscle precursor cells at an early stage when they start to migrate from the dermamyotome of the somites into the limb mesenchyme. Lbx1 knock-out mice show a subdivision of this population of migrating muscle precursor cells in two subpopulations: cells that still have the capacity to migrate and cells that are unable to finish this process. Lbx1 also specifies a subpopulation of cardial neural crest cells which migrate into the tubular heart. The Effect of a loss of Lbx1function is not restricted to migration processes but also affects the heart of mutant mice leadingto hyperplasia of the myocard caused by an increased proliferation rate. During development of interneurons in the neural tube Lbx1 determines a ventral subpopulation of dorsal interneurons and adds positional information to a network of dorsalizing and ventralizing factors. Knock-out mice show a decrease in the number of neurons in this region. Lbx1 knock-out mice and Pax3 mutant mice reveal an involution of both genes in a negatively regulating feed-back loop.
