dc.description.abstractDas Enzym Hexokinase (Hxk) katalysiert die Phosphorylierung von Hexosezuckern am Atom C6 und macht sie somit dem Stoffwechsel verfügbar. Bevorzugte Substrate sind Glukose und als Phosphatdonor ATP. Durch Hybridierungs- und PCR-Techniken wurden im Tabak-Genom (Nicotiana tabacum) zehn verschiedene Hexokinase-Gene nachgewiesen. Sieben von ihnen wurden als cDNA isoliert und als Hxk1 bis Hxk7 veröffentlicht. Die verschiedenen Hexokinase-Isoformen konnten in Übereinstimmung mit anderen pflanzlichen Hexokinasen in die Typen A, B1 und B2 gruppiert werden. Hxk2 ist ein Typ A Enzym und im Plastidenstroma lokalisiert. Ihr Promotor zeigte Aktivität in Xylem-Parenchym und Stärkescheide des Leitgewebes, in Schließzellen und Wurzelspitzen. Die anderen Isoformen gehören dem Typ B1 (Hxk6) bzw. Typ B2 (restliche) an. Ihre Fusionen mit GFP waren mit Mitochondrien assoziiert, wo sie vermutlich auf der zytosolischen Seite in der äußeren Hüllmembran verankert sind. Der Promotor von Hxk7 wurde aus Nicotiana sylvestris isoliert und war fast ausschließlich in Pollen aktiv. Für Hxk1 ist die gleiche Gewebespezifität anzunehmen. Hxk3-Transkripte akkumulierten am stärksten in Staubbeuteln, jedoch nicht so dominant wie Hxk1 und Hxk7. Die Gewebespezifität von Hxk4, Hxk5 und Hxk6 konnte nicht geklärt werden. Nach Überexpression in Tabak wurden pH-Abhängigkeit und kinetische Eigenschaften der Isoformen 1-6 bestimmt. Hxk6 zeigte keine katalytische Aktivität, verursachte jedoch geringfügig gebleichte Sink-Bereiche junger Blätter. Die biochemischen Eigenschaften der anderen Isoformen lagen im üblichen Bereich pflanzlicher Hexokinasen. Auffällig war die durchweg geringste Substrat-Affinität von Hxk2, sowie die sehr hohe Affinität von Hxk4 und Hxk5 zu ATP und Fruktose. Aus den Einzelergebnissen wurden Modelle für die Funktion der jeweiligen Isoform innerhalb der Pflanze abgeleitet.-
dc.description.abstractThe enzyme hexokinase (Hxk) catalyzes the phosphorylation of hexose sugars at atom C6 and thus makes them accessible for further metabolism. Preferred substrates are glucose and as phosphate donor ATP. In the Nicotiana tabacum genome 10 different hexokinase genes could be detected, seven of which were isolated as cDNA and published as Hxk1 - Hxk7. In accordance with other plant hexokinases the tobacco enzymes could be grouped into the three types A, B1 and B2. Hxk2 is a type A enzym and localized in the plastid stroma. The promoter was active in the xylem parenchyma and starch sheath of the vascular bundles, in guard cells and root tips. The other isoforms belong to type B1 (Hxk6) and type B2 (all others). By fusion to GFP they were found associated with mitochondria, most probably attached to the cytosolic side of the outer membrane. The promoter of Hxk7 was isolated from Nicotiana sylvestris and showed activity almost exclusivley in pollen. It is assumed that Hxk1 has the same tissue specificity. Transcripts of Hxk3 accumulated in anthers mostly, but not as predominatly as those of Hxk1 and Hxk7. Tissue specificities of Hxk4, Hxk5 and Hxk6 remain unknown. Hxk1 - Hxk6 were overexpressed in tobacco for determination of their biochemical properties. Hxk6 showed no catalytic activity but led to pale sink areas of young leaves. Properties of the other isoforms were in the range of already published plant hexokinases. Noticeable were the relatively low substrate affinities of Hxk2 and the outstanding high affinities of Hxk4 and Hxk5 to ATP and fructose. Based upon the experimental results individual models were built for the roles of the different hexokinase isoforms within the plant metabolism.eng
