Abstract: Calystegine sind Nortropanalkaloide mit Glycosidase-hemmender Aktivität. Solanum dulcamara L. zeigte ein besonderes Muster an Calysteginen. Calystegin A3 dominiert das Calystegingemisch, was ungewöhnlich für Solanaceen ist, da normalerweise B2 das vorherrschende Calystegin darstellt. Der höchste Gehalt an Gesamtcalysteginen bezogen auf Trockenmasse wurde in Sprossspitzen und Blüten gefunden. Tropinonreduktasen sind als kurzkettige Dehydrogenesen/Reduktasen (SDR) Bestandteil der Tropanalkaloidbiosynthese. In Solanum dulcamara L. ist eine pseudotropinformende Tropinonreduktase (TRII) an der Biosynthese der Calystegine beteiligt. Eine TRII aus S. dulcamara wurde kloniert und heterolog in E. coli exprimiert. Das Protein wurde mit Histidinrest gereinigt. Die ungewöhnlichen kinetischen Parameter der Tropinonreduktase führten zu der Frage nach der Wirkung des Histidinrestes auf die katalytischen Eigenschaften. Die Proteinstruktur wurde modelliert und erklärte, wie der Histidinrest am C-Terminus jedoch nicht am N-Terminus einen Einfluss auf die Katalyse ausüben kann. Mit dem Ziel neue Liganden und potentiellen Inhibitoren der SdTRII zu finden, wurde eine Pharmakophor basierte Suche in zwei Datenbanken durchgeführt. Die in der Suche identifizierten Strukturen wurden in das aktive Zentrum des Tropinonreduktase-Modells mit einem Ligand Docking Programm eingepasst und entsprechend der Fitness ausgewertet. Strukturen mit der besten Bewertung wurden als Substrate und Inhibitoren für in vitro Tests verwendet. Neue Liganden mit inhibitorischer Wirkung auf eine SDR der Alkaloidbiosynthese wurden identifiziert.
Calystegines are nortropane alkaloids with glycosidase inhibitory activities. Solanum dulcamara L., proved to possess a special pattern of calystegines. Calystegine A3 dominated the calystegine mixture, which is uncommon for Solanaceae; the prevailing compound usually is calystegine B2. The highest contents of total calystegines per dry mass were found in shoot tips and flowers. Tropinone reductases are short-chain dehydrogenases/oxidases (SDR) involved in tropane alkaloid biosynthesis. In Solanum dulcamara L., a tropinone reductase specific for pseudotropine formation (TRII) participates in the biosynthesis of calystegines. TRII from S. dulcamara was cloned and heterologously expressed in E. coli. The protein was purified using the hexahistidine-tag. Unusual kinetic characteristics of this tropinone reductase II prompted to investigate a possible impact of the hexahistidine affinity tag on catalytic properties. Protein structure was modeled and explained how the His-tag at the C-terminus but not at the N-terminus can influence the catalysis. In order to find novel ligands and potential inhibitors for tropinone reductases, a pharmacophore-guided screening through chemical structure databases was performed. Structure hits identified in the screening were fitted into the active site of a tropinone reductase model by a ligand docking program and ranked according to their resulting fitness scores. Structures with best scores were tested in vitro as substrates and inhibitors. Novel ligands with inhibitory potential for a SDR of alkaloid biosynthesis were identified.
