Abstract: Das Typ-III-Effektorprotein AvrBs3 aus Xanthomonas campestris pv. vesicatoria wird über das Typ-III-Sekretionssystem in die Pflanzenzelle transloziert. AvrBs3 wird in den Kern der Pflanzenzelle importiert, induziert die Transkription von pflanzlichen Genen (upa, "upregulated by AvrBs3") und löst eine Hypertrophie, d.h. Vergrößerung, von Mesophyllzellen aus. Im Rahmen dieser Arbeit wurden 12 neue upa-Gene mittels subtraktiver Hybridisierung aus Paprika isoliert. upa20, das einen Transkriptionsfaktor der bHLH-Familie kodiert, erwies sich als notwendig und hinreichend für die Auslösung der Hypertrophie in verschiedenen Solanaceen. Upa20 induziert die Transkription zweier in früheren Arbeiten isolierter upa-Gene, die ein α-Expansin bzw. einen Transkriptionsfaktor kodieren. Mittels β-Glucuronidase als Reporter wurde die AvrBs3-responsive Region des upa20-Promotors auf 55 bp eingegrenzt. Der Sequenzvergleich mit anderen upa-Promotoren führte zur Identifizierung der upa-Box, eines ca. 20 bp langen DNA-Motivs, das die AvrBs3-abhängige Aktivierung von Genen vermittelt. Dieses Motiv wurde durch upa20-Promotormutanten näher charakterisiert. AvrBs3 nutzt einen alternativen Transkriptionsstart, der stromabwärts des für die Basalexpression hauptsächlich genutzten Startpunkts lokalisiert ist. Die upa-Box befindet sich innerhalb von 100 bp stromaufwärts des Transkriptionsstarts, der in Anwesenheit von AvrBs3 verwendet wird. Die Gene upa14 und upa19 werden auch durch das AvrBs3-Derivat AvrBs3Δrep16 induziert, dem vier der 17,5 zentra len Sequenzwiederholungen in AvrBs3 fehlen. AvrBs3Δrep16 erkennt eine mutierte Version der upa-Box im upa20-Promotor. Um für zukünftige Analysen der AvrBs3-Familie auch die Modellpflanze Arabidopsis thaliana bzw. verwandte Spezies nutzen zu können, wurden im zweiten Teil der vorliegenden Arbeit die AvrBs3-Homologen Hax2, Hax3 und Hax4 aus dem Brassicaceen-Pathogen X. campestris pv. armoraciae analysiert. Ihre Typ-III-abhängige Translokation in die Pflanzenzelle wurde über Domänenaustauschexperimente mit AvrBs3 gezeigt. Alle drei hax-Gene tragen zur Ausbildung Krankheits-assoziierter Chlorosen und Nekrosen in Radieschenpflanzen bei, wobei hax2 den stärksten Einfluss auf die Symptombildung hatte. Weiterhin löste hax2 bei transienter Expression in Arabidopsis thaliana Col-0 eine Akkumulation von Anthocyanen aus. Hax3 und Hax4, aber nicht Hax2, zeigen außerdem Avirulenzaktivität in Tomate, wo sie durch das Resistenzprotein Bs4 erkannt werden.
The type III effector protein AvrBs3 from Xanthomonas campestris pv. vesicatoria is translocated into the plant cell by the type III secretion system. AvrBs3 is imported into the plant cell nucleus, induces the transcription of plant genes (upa, upregulated by AvrBs3) and elicits a hypertrophy, i.e. enlargement, of mesophyll cells in several solanaceous plants. In this work, 12 new upa genes were isolated from pepper by subtractive hybridization. upa20 encodes a transcription factor of the basic helix-loop-helix family and was necessary and sufficient for the elicitation of hypertrophy in several solanaceous plants. Upa20 induces the transcription of two previously isolated upa genes encoding an a-expansin and a transcription factor, respectively. Using ß-glucuronidase as a reporter the AvrBs3-responsive region in the upa20 promoter could be restricted to 55 bp. Sequence comparisons with other upa promoters led to the identification of the upa box, a DNA motif of approximately 20 bp, which mediates AvrBs3-dependent gene activation. This motif was characterized further by upa20 promoter mutants. AvrBs3 uses an alternative transcription initiation site which is located upstream of the transcription start generally used for basal expression. The upa-Box is localized within 100 bp upstream of the transcription start used in the presence of Avrbs3. The genes upa14 and upa19 are induced also by the AvrBs3 derivative AvrBs3?rep16 which lacks four out of the 17.5 central repeats in AvrBs3. AvrBs3?rep16 recognizes a mutated version of the upa box in the upa20 promoter. To allow the use of the model plant Arabidopsis thaliana and related species in future analyses of the AvrBs3 family, in the second part of this work the AvrBs3 homologs Hax2, Hax3 and Hax4 from the crucifer pathogen X. campestris pv. armoraciae were analyzed. Their type III-dependent translocation into the plant cell was shown by domain swap experiments. All three hax genes contribute to the formation of disease-associated chlorosis and necrosis in radish plants with hax2 having the strongest influence on symptom formation. In addition, transiently expressed hax2 caused an accumulation of anthocyanins in Arabidopsis thaliana Col-0. Hax3 and Hax4, but not Hax2, show avirulence activity in tomato where they are recognized by the resistance protein Bs4.
