dc.description.abstract | Das durch das PAMP Pep-13 induzierte Solanum tuberosum-Gen StIPP kodiert für ein Enzym, das in vitro spezifisch Phosphatidylinositol-4,5-bisphosphat (PI(4,5)P2) zu Phosphatidylinositol-4-monophosphat (PI4P) dephosphorylierte. Beide Phosphoinositide sind an der Regulation exo- und endozytotischer Prozesse an der Plasmamembran (PM) beteiligt. Transient exprimiertes StIPP-GFP-Protein lokalisierte in nicht infizierten Nicotiana benthamiana-Zellen an der PM und nach Infektion mit dem Pathogen Phytophthora. infestans zusätzlich an der extrahaustoriellen Membran. Die transiente StIPP-Expression in Arabidopsis thaliana-Protoplasten führte zur erhöhten Abundanz eines PAMP-Rezeptors. Obwohl eine veränderte StIPP-Expression in S. tuberosum-Transformanten keine Auswirkungen auf die Suszeptibilität oder Resistenz gegenüber P. infestans hatte, deuten die Ergebnisse dieser Arbeit insgesamt auf eine Funktion innerhalb PI(4,5)P2- oder PI4P-abhängier Prozesse während der Pathogen-Wirts-Interaktion. | ger |
dc.description.abstract | The Solanum tuberosum gene StIPP is induced by the pathogen-associated molecular pattern (PAMP) Pep-13 The gene encodes an enzyme that specifically dephosphorylates phosphatidylinositol 4,5-bisphosphate (PI(4,5)P2) to phosphatidylinositol 4-monophosphate (PI4P) in vitro. Both phosphoinositides are involved in the regulation of exo- and endocytic processes at the plasma membrane. When transiently expressed in Nicotiana benthamiana, StIPP-GFP localizes to the plasma membrane in non-infected cells and additionally to the extrahaustorial membrane after infection with the pathogen Phytophthora infestans. StIPP expression in Arabidopsis thaliana protoplasts resulted in the stabilization of a PAMP receptor. Although altered StIPP expression in transgenic S. tuberosum lines had no effect on susceptibility or resistance to P. infestans, the overall results of this work suggest a function within PI(4,5)P2- or PI4P-dependent processes during pathogen-host interaction. | eng |
