Abstract: In dieser Arbeit werden die drei myxobakteriellen Substanzen Argyrin C, Aurachin D und HZI 15 beschrieben, die einen c-FLIPL-modulierenden Effekt aufweisen. Der Apoptosesensitivierende Effekt dieser Wirkstoffe ist ein erster Schritt zur Entwicklung von Medikamenten, die etwa in der Tumormedizin, Transplantationsmedizin oder gegen Infektionen Anwendung finden können. Apoptose ist ein elementarer Prozess in Eukaryoten. Sie spielt unter anderem eine Rolle in der Embryonalentwicklung, der Zellhomöostase des Immunsystems und der Eliminierung von entarteten oder infizierten Zellen. Dementsprechend hat eine deregulierte Apoptose gravierende Auswirkungen auf den jeweiligen Organismus und ist ein bedeutender Mechanismus in vielen Erkrankungen. Ein wichtiges Apoptose-regulierendes Protein ist c-FLIP. Seine Rolle in Infektionen, Autoimmunerkrankungen und der Krebsentstehung wurden schon eingehend untersucht. Zudem sind auch andere klinisch relevante Prozesse wie Transplantationen auf die Regulierung von c-FLIP angewiesen, da die Entwicklung von T- und B-Zellen, sowie von regulatorischen T-Zellen, von diesem Protein abhängig ist. Trotz des offensichtlichen Potentials c-FLIPmodulierender Medikamente ist noch keine Substanz bekannt, die direkt auf c-FLIP wirkt. Ziel dieser Arbeit war es daher, c-FLIP-modulierende Wirkstoffe zu finden und diese zu charakterisieren. Die gefundenen Substanzen wurden auf eine mögliche Eignung als Medikament getestet und die ersten Experimente auf dem Weg zu klinischen Tests wurden mit ausgewählten Wirkstoffen durchgef¨uhrt. Dazu wurde eine Bibliothek von 259 myxobakteriellen Substanzen auf ihre Wirkung auf c-FLIP analysiert. Dreizehn dieser Substanzen konnten c-FLIPL- überexprimierende Zellen zusammen mit dem Todesliganden CD95L auf den Zelltod sensitivieren. Bei Nachtests mit elf Wirkstoffen konnte gezeigt werden, dass diese Substanzen die Zellen zur Apoptose stimulieren. Die drei Wirkstoffe Argyrin C, Aurachin D und HZI 15 wurden für eine nähere Charakterisierung ausgewählt. Alle drei Substanzen sensitivieren c-FLIPL- überexprimierende Zellen für Apoptose. Argyrin C wirkt bis zu einer Konzentration von 10 μM, wobei bei einer Konzentration von 25 μM toxische Effekte sichtbar sind. Aurachin D wirkt ebenfalls bis zu einer Konzentration von 10 μM. Ein leichter toxischer Effekt ist bei dieser Substanz nur im Langzeittest mit Leervektorzellen sichtbar, da hier die Viabilität unter Aurachin D-Einfluss nach 28 h beständig absinkt. Die in der niedrigsten Konzentration wirksame Substanz in dieser Arbeit ist HZI 15. Bei dieser konnten Effekte bis zur geringsten getesteten Konzentration von 10 nM beobachtet werden. HZI 15 zeigt daf¨ur bei höheren Konzentrationen ab etwa 10 μM toxische Effekte. Alle Substanzen zeigen nur bei einem funktionellen extrinsischen Apoptosesignalweg eine Apoptose-sensitivierende Wirkung. Das mRNA-Level von FLIP sinkt unter dem Einfluss der drei genauer charakterisierten Substanzen nach 16 h nicht ab. Argyrin C hat kaum einen Effekt auf die Proteinexpression von c-FLIPL, wobei die Expression von c-FLIPS verringert ist. Aurachin D verringert die Expression von c-FLIPL und c-FLIPS. Unter HZI 15- Einfluss ist nach 24 h eine deutlich stärkere Verringerung der c-FLIPL-Expression in c-FLIPLZellen sichtbar als nach 16 h. Schon nach 16 h ist die Expression von c-FLIPS in c-FLIPS- überexprimierenden Zellen stark verringert.
In this thesis, three myxobacterial compounds, Argyrin C, Aurachin D and HZI 15, are described, that show a c-FLIP modulating effect. The apoptosis-sensitising effect of these substances is a first step towards the development of drugs, that can be used in a wide range in the clinic, including tumor medicine, transplantation medicine or against infections. Apoptosis is a fundamental process in eukaryotic organisms. It plays a role in the embryonic development, the homeostasis of the immune system and the elimination of mutated or infected cells. Therefore deregulated apoptosis has a serious impact on the affected organism and is an important mechanism in a number of diseases. A major apoptotic-regulating protein is c-FLIP. Its role in infections, autoimmune-diseases and development of cancer has been studied intensively. Also other clinical relevant procedures like transplantations are dependent on the proper regulation of c-FLIP as some cell populations like T- and B-cells as well as Tregs are dependent on this protein. Despite the obvious potential of c-FLIP modulating drugs, no substances are known, yet, that directly interact with c-FLIP. So the aim of this thesis was to find and characterise c-FLIP modulating compounds. The respective substances were investigated for their suitability as a clinical drug and the first steps to clinical testings were performed with selected compounds. A library of 259 myxobacterial compounds were screened for their effect on c-FLIP. Thirteen of these substances could sensitise c-FLIPL-overexpressing cells in combination with the death ligand CD95L to cell death. Additional tests with eleven of these thirteen compounds showed that all of the tested substances indeed induce apoptosis. The three compounds Argyrin C, Aurachin D and HZI 15 were chosen for further characterisation. All of these three substances sensitise c-FLIPL-overexpressing cells to apoptosis. Argyrin C shows an activity up to a concentration of 10 μM, with toxic effects at a concentration of 25 μM. Aurachin D was also active up to a concentration of 10 μM. A slight toxic effect was visible in long-time experiments on empty vector cells. Here, the viability of the cells with Aurachin D treatment dropped steadily after 28 h. The most active compound in this screening was HZI 15, showing effects even in the lowest tested concentration of 10 nM. But HZI 15 also showed toxic effects at higher concentrations starting at approximately 10 μM. All of the three tested compounds only show the apoptosis-sensitizing effect when tested on cells with a functional extrinsic apoptosis pathway. Interestingly, the mRNA level did not decrease when the cells were treated with the three substances for 16 h. Argyrin C only showed a slight effect on the protein expression of c-FLIPL but c-FLIPS-expression was decreased. Aurachin D reduced the expression of both, c-FLIPL and c-FLIPS. When treated with HZI 15, less c-FLIPL was detected after 24 h of treatment. The c-FLIPS-expression in c-FLIPS-expressing cells was decreased already after 16 h of treatment.
