dc.description.abstract | In this work, all known and essential protein complexes of the mRNA 3’ processing (CFI, CFII, CstF, mPSF, and the endonuclease complex) were recombinantly expressed and chromatographically purified. For structural analysis, these complexes were cross-linked with the DSBU cross-linker and analyzed by mass spectrometry. Additionally, the recombinant protein complexes were used as bait proteins to identify interaction partners in nuclear extract. | eng |
dc.description.abstract | In dieser Arbeit wurden alle bekannten und essentiellen Proteinkomplexe der mRNA-3‘-Prozessierung (CFI, CFII, CstF, mPSF und der Endonukleasekomplex) rekombinant exprimiert und chromatografisch gereinigt. Zur strukturellen Analyse wurden diese Komplexe mit dem Cross-Linker DSBU chemisch vernetzt und massenspektrometrisch analysiert. Zusätzlich wurden die rekombinanten Proteinkomplexe als Köderprotein eingesetzt, um Interaktionspartner in Kernextrakt zu identifizieren. | ger |
